目的探讨紧密连接蛋白CLDN1和CLDN4在人肝细胞肝癌(human hepatocellular carcinoma,HCC)组织中的表达及其意义。方法郑州大学第一附属医院2010年至2011年手术切除的HCC组织标本60例和正常肝组织50例,采用免疫组织化学二步法分别检测CLDN1和CLDN4的表达。结果 CLDN1和CLDN4在HCC组织中的阳性表达率分别为70%(42/60)和25%(15/60),两者的表达与患者年龄、性别、血液AFP浓度无关(P>0.05),与肿瘤分化程度无关(P>0.05)。CLDN1和CLDN4在HCC中的表达具有相关性(r=0.533,P<0.05)。CLDN1在正常肝组织中无表达(0/50),CLDN4在正常肝组织中高表达(50/50)。两者在正常肝组织中的表达具有明显差异(P<0.05)。结论肝细胞肝癌中存在CLDN1和CLDN4的异常表达,CLDN1在HCC中表达上调,而CLDN4在HCC中表达下调,两者表达具有相关性(r=0.533,P<0.05)。推测CLDN1表达上调和CLDN4表达下调能够促进肝细胞肝癌的发生发展。
Objective To investigate the expression and significance of the claudin CLDN1 and CLDN4 in human hepatocellular carcinoma (HCC). Methods Sixty cases of HCC tissues and 50 cases of normal liver tissues were collected from the First Affiliated Hospital of Zhengzhou University from 2010 to 2011. The expression of CLDN1 and CLDN4 were detected by immunohistochemical two-step method. Results The positive rates of CLDN1 and CLDN4 in HCC tissues were 70% (42/60) and 25% (15/60), respectively. The expressions of CLDN1 and CLDN4 were not related to the age, sex and blood AFP concentration (P> 0.05) No correlation with tumor differentiation (P> 0.05). There was a correlation between CLDN1 and CLDN4 expression in HCC (r = 0.533, P <0.05). CLDN1 was not expressed in normal liver tissue (0/50), and CLDN4 was overexpressed (50/50) in normal liver tissue. There was a significant difference between the two in normal liver tissue (P <0.05). Conclusions The abnormal expression of CLDN1 and CLDN4 in hepatocellular carcinoma is observed. The expression of CLDN1 is up-regulated in HCC, while the expression of CLDN4 is down-regulated in HCC (r = 0.533, P <0.05). It is speculated that CLDN1 upregulation and CLDN4 downregulation can promote the occurrence and development of hepatocellular carcinoma.
