目的:观察抑郁大鼠模型脑内环磷酸腺苷(cAMP)通路的表达及中药颐脑解郁方的干预作用。方法:Wistar大鼠分为正常组、模型组、中药颐脑解郁方组、西药氟西汀组。模型组、中药颐脑解郁方组、西药氟西汀组在21 d内采用慢性不可预见的温和性应激加孤养法(method of chronic unpredictable mild stress with isolated support)建立抑郁状态大鼠模型,模型组ig双蒸水,中药治疗组、西药对照组分别ig中药颐脑解郁方(6.2 g.kg-1)、西药氟西汀(剂量2.33 mg.kg-1),采用RT-PCR实验方法,测定PDE4(3’,5’-cyclic-nucleotide phosphodiesterase)、CREB(cAMP response element binding protein)在抑郁模型大鼠脑内左前额叶皮质、海马的表达。结果:模型组大鼠大脑额叶皮质及海马PDE4表达增强(P<0.05),CREB表达明显低于正常组(P<0.05,P<0.01);中药颐脑解郁方及西药氟西汀干预后,额叶前侧皮质及海马PDE4的表达较模型组有明显的降低(P<0.05,P<0.01)。CREB表达明显增高(P<0.05,P<0.01)。结论:中药颐脑解郁方干预后可明显调节PDE4及CREB在抑郁大鼠脑内的表达,提示中药颐脑解郁方的抗抑郁作用可能与调节cAMP信号转导通路有关。
Objective: To observe the expression of cyclic adenosine monophosphate (cAMP) pathway in the brain of depressive rats and the intervention effect of Chinese herbal medicine Yinao Jieyu Decoction. Methods: Wistar rats were divided into normal group, model group, Chinese medicine Yinao Jieyu Fang group and western medicine fluoxetine group. The model group, the traditional Chinese medicine Yinao Jieyu Recipe group and the western medicine fluoxetine group were used to establish the depressive state rat model within 21 days by the model of chronic unpredictable mild stress with isolated support Group ig double distilled water, traditional Chinese medicine treatment group, Western medicine control group were ig Chinese medicine Yinao Jieyu Decoction (6.2 g.kg-1), Western medicine fluoxetine (2.33 mg.kg-1 dose), using RT-PCR test method PDE4 (3 ’, 5’-cyclic-nucleotide phosphodiesterase) and CREB (cAMP response element binding protein) in the left frontal cortex and hippocampus of the depression model rat brain. Results: The expression of PDE4 in the frontal cortex and hippocampus in the model group was increased (P <0.05), and the expression of CREB was significantly lower in the model group than in the normal group (P <0.05, P <0.01). After the Chinese medicine Yinao Jieyu Recipe and western medicine fluoxetine were interfered, The expression of PDE4 in the frontal cortex and hippocampus was significantly lower than that in the model group (P <0.05, P <0.01). CREB expression was significantly increased (P <0.05, P <0.01). Conclusion: The Chinese herbal medicine Yinao Jieyu Decoction can significantly regulate the expression of PDE4 and CREB in the brain of depression rats, suggesting that the anti-depression effect of Yinao Jieji Prescription may be related to the regulation of cAMP signal transduction pathway.
