脂质体作为药物载体的试验很多,某些已获得成功,特别是以药物输送至网状内皮细胞作为抗寄生虫疾病的治疗、酶治疗、金属螯合治疗,以及抗癌转移治疗。脂质体还可用为药物贮存系统,持续释放药物至有利的部位。用脂质体包裹药物(例如二性霉素B)可以减低药物毒性。脂质体工程中的最新进展如单克隆抗体的共价联结和pH敏感及热敏感的脂质体的设计,可以扩大其应用范围。
There are many experiments with liposomes as drug carriers, some of which have been successful, particularly drug delivery to reticuloendothelial cells as an antiparasitic disease treatment, enzymatic treatment, metal chelation therapy, and anti-cancer metastasis therapy. Liposomes can also be used as drug storage systems to deliver sustained release of drug to a favorable site. Drug-encapsulation of liposomes (eg, amphotericin B) can reduce drug toxicity. Recent advances in liposome engineering such as the covalent attachment of monoclonal antibodies and the design of pH-sensitive and heat-sensitive liposomes can broaden their range of applications.
