目的 研究转IL—2基因对人肺腺癌细胞(SPC—A_1)药物敏感性的影响。方法 用MTT法检测阿霉素(ADM)、丝裂霉素C(MMC)、顺铂(DDP)、5—氟脲嘧啶(5—Fu)对导入IL—2基因的肺腺癌细胞株(SPC—A_1/IL—2)的杀伤率,并与未转染IL—2基因的人肺腺癌细胞株(SPC—A_1)比较。结果 ADM、MMC、DDP、5—Fu对SPC—A1/IL—2的杀伤率均高于SPC—A_1,但ADM、MMC、DDP对SPC—A_1/IL—2的半数致死量(LD_(50))与其对SPC—A_1的LD_(50)比较,没有统计学意义(t′=0.175,P>0.05)。而5>Fu对SPC—A_1/IL—2的LD_(50)为23.50±3.04μg/ml,对SPC—A_1的LD_(50)则为39.47±4.01μg/ml,两组LD_(50)比较差异显著(t′=10.58,P<0.001)。结论 转IL—2基因的人肺腺癌细胞对ADM、MMC、DDP的敏感性没有改变,但增加了对5—Fu的敏感性。
Objective To study the effect of transfected IL-2 gene on the sensitivity of human lung adenocarcinoma cells (SPC-A_1). Methods MTT assay was used to detect adriamycin (ADM), mitomycin C (MMC), cisplatin (DDP) and 5-fluorouracil (5-Fu) against lung adenocarcinoma cell lines that introduced IL-2 gene ( The killing rate of SPC-A_1/IL-2 was compared with that of human lung adenocarcinoma cell line (SPC-A_1) which was not transfected with IL-2 gene. Results The killing rate of ADM, MMC, DDP, 5-Fu on SPC-A1/IL-2 was higher than that of SPC-A_1, but the median lethal dose of ADM, MMC, and DDP on SPC-A_1/IL-2 (LD_(50) Compared with LD_(50) to SPC-A_1, there was no statistical significance (t′=0.175, P>0.05).
