目的研究积雪草苷对心肌梗死(心梗)大鼠左室心肌纤维化和心肌组织转化生长因子-β1(transforming growth factor-β1,TGF-β1)、胶原Ⅰ(collagenⅠ,ColⅠ)和胶原Ⅲ(collagenⅢ,ColⅢ)表达的影响。方法结扎雄性SD大鼠左冠状动脉前降支建立心梗模型,随机分为假手术组(Sham组)、假手术+积雪草苷组(Sham+Asiaticoside组)、心梗模型组(MI组)和心梗模型+积雪草苷组(MI+Asiaticoside组)。造模当天给药,早晚各1次,2周后分别测定各组大鼠心脏左室重量与体重之比、左室梗死面积;免疫组织化学(免疫组化)检测心肌组织非梗死区TGF-β1、ColⅠ、ColⅢ的表达。计量资料采用t检验,P<0.05为差异有统计学意义。结果 MI组左室梗死面积为(24.37±5.69)%,MI+Asiaticoside组左室梗死面积为(25.89±6.27)%,差异无统计学意义(P>0.05);MI组左室重/体重为(3.46±0.26)mg/g,MI+Asiaticoside组左室重/体重为(2.53±0.35)mg/g,差异有统计学意义(P<0.05);免疫组化染色结果显示,与MI组相比[(8.34±0.62)、(37.66±2.21)、(12.39±1.37)μm2],MI+Asiaticoside组非梗死区心肌TGF-β1、ColⅠ、ColⅢ的表达[(5.97±0.78)、(17.90±1.36)、(8.14±1.23)μm2]明显降低,差异均有统计学意义(均P<0.05)。心梗大鼠非梗死区心肌组织ColⅠ、ColⅢ表达与心肌组织TGF-β1的表达正相关。结论积雪草苷可减少心梗大鼠非梗死区TGF-β1的表达,从而减弱非梗死区心肌组织ColⅠ、ColⅢ异常合成与沉积,减轻心梗后左室心肌纤维化程度,延缓左室重构的发展。
Objective To study the effects of asiaticoside on left ventricular fibrosis and the expression of transforming growth factor-β1 (TGF-β1), collagen Ⅰ (ColⅠ) and collagen Ⅲ in myocardial infarction (collagen Ⅲ, Col Ⅲ) expression. Methods The left anterior descending coronary artery of male Sprague-Dawley rats was ligated to establish a myocardial infarction model. The expression of ColⅠ and Col Ⅲ in non-infarcted myocardium of myocardial infarction rats was positively correlated with the expression of TGF-β1 in myocardium. Conclusion Asiaticoside can reduce the expression of TGF-β1 in non-infarcted area of myocardial infarction rats, and attenuate the abnormal synthesis and deposition of ColⅠ and Col Ⅲ in non-infarcted myocardium, and reduce the degree of left ventricular fibrosis after myocardial infarction, The development of architecture.
