目的 :探讨细胞周期素依赖性激酶 2 (Cdk2 )及其抑制因子p2 7Kip1与胃癌生物学行为的关系。方法 :采用免疫组织化学S P法对 6 0例胃癌组织原发灶和淋巴结转移灶进行Cdk2、p2 7Kip1免疫组化染色 ,腹水或腹腔冲洗液行脱落细胞学检查。结果 :Cdk2阳性表达率 5 6 .6 7% (34/ 6 0 ) ,阳性细胞定位于细胞核和细胞浆 ,以Ⅲ、Ⅳ期表达率较高 (P <0 .0 1) ,与胃癌大体类型及浆膜分型关系密切 (P <0 .0 1) ;p2 7Kip1阳性表达率为 4 3.33% (2 6 / 6 0 ) ,阳性细胞定位于细胞浆 ,在高分化及Ⅰ、Ⅱ期胃癌高表达 (P <0 .0 5 ) ,且与淋巴结转移密切相关 (P <0 .0 1) ;Cdk2、p2 7Kip1同时阳性表达 19例 ,占 31.6 7% (19/ 6 0 )。结论 :Cdk2与胃癌浸润深度、淋巴结转移呈正相关 ,p2 7Kip1表达与胃癌浸润深度、淋巴结转移呈负相关
Objective: To investigate the relationship between cyclin dependent kinase 2 (Cdk2) and its inhibitor p27Kip1 and the biological behavior of gastric cancer. Methods: Sixty cases of primary gastric cancer and lymph node metastasis were immunohistochemically stained with Cdk2 and p27Kip1 immunohistochemical staining, and ascites or peritoneal lavage fluid were exfoliated cytologically. Results: The positive rate of Cdk2 was 56.7% (34/60). The positive cells were located in nucleus and cytoplasm. The positive rate of Cdk2 was higher in stage Ⅲ and Ⅳ (P <0.01) (P <0.01). The positive expression rate of p27Kip1 was 43.33% (26/60). The positive cells located in the cytoplasm. In the highly differentiated and high grade Ⅰ and Ⅱ gastric cancer (P <0.05), which was closely related to lymph node metastasis (P <0.01). Meanwhile, 19 cases (31.6%) were positive for Cdk2 and p27Kip1, accounting for 31.6% (19/60). Conclusion: Cdk2 is positively correlated with the depth of invasion and lymph node metastasis, and the expression of p27Kip1 is negatively correlated with the depth of invasion and lymph node metastasis
