目的 研究泰素 (Taxol)结合放射对人鼻咽癌细胞系 (CNE)的联合作用效应。方法 用克隆形成法制作细胞存活曲线 ,流式细胞分析测定细胞周期分布。Tunel法作细胞凋亡测定。结果 不同浓度的Taxol与CNE细胞作用 2 4h ,其细胞毒性呈剂量依赖性。在放射增敏实验中 ,10 0nmol/LTaxol作用 2 4h ,对CNE细胞有放射增敏作用 ,增敏比为 1.2 3。同样剂量的Taxol作用同样时间 ,可诱导CNE细胞的G2 +M期阻滞。Tunel法测定细胞凋亡 ,10 μmol/LTaxol作用 2 4h ,可诱导CNE细胞凋亡 ,X射线照射 2Gy也可诱导CNE细胞凋亡 ,而 10 μmol/LTaxol与 2GyX射线同时作用 ,诱导凋亡明显增加。结论 Taxol对CNE细胞有放射增敏作用 ,除自身对细胞凋亡有诱导作用外 ,还能增强放射对CNE细胞凋亡的诱导作用。
Objective To investigate the combined effects of Taxol combined with radiotherapy on human nasopharyngeal carcinoma cell line (CNE). Methods Cell survival curves were generated by clonogenic assay and cell cycle distribution was determined by flow cytometry. Tunel method for determination of apoptosis. Results Different concentrations of Taxol and CNE cells for 24 hours, the cytotoxicity in a dose-dependent manner. In radiosensitization experiments, 10 0 nmol / LTaxol for 24 h had a radiosensitizing effect on CNE cells with a sensitization ratio of 1.2 3. The same dose of Taxol the same time, can induce CNE cells G2 + M phase arrest. Tunel assay of apoptosis, 10μmol / LTaxol effect 24h, can induce apoptosis of CNE cells, X-ray irradiation 2Gy also induced CNE cell apoptosis, while 10μmol / LTaxol and 2Gy X-rays at the same time, induced apoptosis was significantly increased . Conclusion Taxol has a radiosensitizing effect on CNE cells. In addition to its own induction of apoptosis, Taxol also enhances the induction of apoptosis on CNE cells.
