目的检测人卵巢癌组织中前胸腺素α(Prothymosinα,PTMA)的表达、分布,并分析其与肿瘤分级的相关性。方法利用卵巢癌组织微阵列,从2块巢癌组织标本和1块癌旁正常组织标本中共获取172份卵巢癌组织和8份癌旁正常组织,对每份肿瘤组织进行病理分级和确定组织类型。采用Western blot法检测卵巢癌和癌旁正常组织中PTMA蛋白的表达水平;免疫组化法检测卵巢癌组织中PTMA蛋白表达的分布。结果 172份卵巢癌组织标本中,病理分级Ⅰ级22份,Ⅱ级67份,Ⅲ级83份,除4份为子宫内膜腺癌外(均为Ⅱ级),其余均为乳头状腺癌。癌旁正常组织中PTMA的表达量明显低于卵巢癌组织,且差异有统计学意义(P<0.01)。8份癌旁正常组织标本中,5份(62.5%)细胞核染色阳性,多出现在卵巢上皮细胞中,3份染色阴性;172份卵巢癌组织标本中,164份(95.3%)为阳性表达,其中细胞核表达44份,细胞质表达40份,混合表达80份;肿瘤分级与PTMA的分布明显相关(P<0.01),卵巢癌Ⅲ级多为混合型分布;卵巢癌Ⅰ级PTMA蛋白的表达水平明显高于卵巢癌Ⅱ、Ⅲ级,且差异有统计学意义(P<0.01)。结论与癌旁正常组织相比,卵巢癌组织中PTMA蛋白的表达增强,在不同分级的肿瘤组织中,PTMA蛋白的表达分布有变化,为进一步研究其在卵巢癌组织中异常表达的临床意义奠定了基础。
Objective To detect the expression and distribution of prothymosin α (PTMA) in human ovarian cancer and to analyze its correlation with tumor grade. Methods Using ovarian cancer tissue microarray, we obtained 172 ovarian cancer tissues and 8 normal tissues adjacent to normal tissues from 2 specimens of cancer tissue and 1 adjacent normal tissue. Pathological grade and tissue type . Western blot was used to detect the expression of PTMA protein in ovarian cancer and adjacent normal tissues. Immunohistochemistry was used to detect the distribution of PTMA protein in ovarian cancer. RESULTS: Of the 172 ovarian cancer specimens, 22 were grade Ⅰ, 67 were grade Ⅱ and 83 were grade Ⅲ, except 4 were endometrial adenocarcinoma (all grade Ⅱ), and the other were papillary adenocarcinoma . The expression of PTMA in adjacent normal tissues was significantly lower than that in ovarian cancer (P <0.01). Of the 8 normal tissues adjacent to cancer, 5 (62.5%) were positive for nucleus staining, mostly in ovarian epithelial cells, 3 were negative for staining, and 164 (95.3%) were positive for 172 of ovarian cancer tissue samples, There were 44 nucleus and 40 cytoplasm, and the expression of PTMA was 80. The grade of PTMA was closely related to the distribution of PTMA (P <0.01) Higher than ovarian cancer Ⅱ, Ⅲ grade, and the difference was statistically significant (P <0.01). Conclusion The expression of PTMA protein in ovarian cancer tissues is increased compared with that in adjacent normal tissues. The expression of PTMA protein varies in different grades of tumor tissues, which lays a foundation for further study on the clinical significance of PTMA protein expression in ovarian cancer tissues The foundation.
