目的探讨双表法对RSV感染小鼠血清中IL-4以及肺组织中TLR-4动态表达的影响。方法将Wister小鼠随机分为正常组、模型组、解表组(银翘散,YQS)、双表组(银翘散+玉屏风散,YPS)和利巴韦林组(RBV)。于感染前3 d双表组,连续给予玉屏风散(7.5 g·kg·d-1)3 d;除正常组以外,50μl 10 LD50病毒液滴鼻建立呼吸道合胞病毒感染的小鼠RSV肺炎模型。感染后2h,正常组和模型组予以双蒸水灌胃,解表组及双表组给与YPS(10 g·kg·d-1)治疗,连续给药3 d;于末次给药后1、3、5、7 d取材,测定不同时间点小鼠肺指数;采用免疫组织化学方法检测小鼠肺组织中TLR-4蛋白的动态表达;ELISA法测定血清中IL-4的含量。结果 YQS、YPS可明显降低小鼠感染后肺指数(P<0.05);治疗后第3天,与模型组相比,YQS、YPS组小鼠血清中IL-4的含量和肺组织中TLR-4蛋白的表达水平有所降低(P<0.05);第5天后较模型组显著降低(P<0.01)。与YQS相比,YPS治疗效果差异显著(P<0.05)。结论双表法对RSV诱导的病毒性肺炎疗效显著,其可能通过抑制TLR-4的蛋白表达,降低血清中IL-4水平,从而达到减轻RSV感染所致的肺损伤的治疗目的。
Objective To investigate the effects of double-table method on IL-4 in serum of mice infected with RSV and dynamic expression of TLR-4 in lung tissue. METHODS: Wistar mice were randomly divided into normal group, model group, relieving group (Yinqiaosan, YQS), double-group (Yinhuasan + Yupingfengsan, YPS) and ribavirin group (RBV). Three days before the infection, the double-tablet group was continuously given Yupingfengsan (7.5 g·kg·d-1) for 3 days. In addition to the normal group, 50 μl of 10 LD50 virus was injected into the nose to establish respiratory syncytial virus-infected mouse RSV pneumonia. model. On the third day after treatment, compared with the model group, IL-4 levels in serum of YQS and YPS mice and TLR in lung tissue The expression level of 4 protein was decreased (P<0.05), and it was significantly lower than the model group on the 5th day (P<0.01). Compared with YQS, the effect of YPS treatment was significantly different (P<0.05). Conclusion The double-table method has a significant effect on RSV-induced viral pneumonia, which may reduce the level of IL-4 in serum by inhibiting the protein expression of TLR-4, so as to reduce the treatment of lung injury caused by RSV infection.
