目的探讨尾加压素Ⅱ(urotensin-2,UTS-Ⅱ)基因rs228651单核苷酸多态性与妊娠糖尿病(gestational diabetes mellitus,GDM)遗传易感性的关系。方法 GDM患者120例(GDM组),正常糖耐量孕妇100例(对照组),采用限制性片段长度多态性PCR法检测2组UTS-Ⅱ基因rs228651位点基因分型,分析UTS-Ⅱ基因rs228651多态性与GDM遗传易感性的关系。结果 GDM组空腹血糖、空腹胰岛素、总胆固醇、胰岛素抵抗指数、β细胞功能指数分别为(6.05±0.51)mmol/L、14.20(13.22,15.60)mu/L,5.85(5.50,6.18)mmol/L,3.82±0.47,116.55(94.19,135.79),对照组分别为(4.40±0.40)mmol/L、10.19(9.63,10.70)mu/L,4.70(4.50,4.90)mmol/L、2.90±0.24、225.56(174.48,314.64),2组比较差异有统计学意义(P<0.05);GDM组UTS-Ⅱ基因rs228651位点GG、AA基因亚型分布频率(44.2%、15.0%)与对照组(23.0%、29.0%)比较差异有统计学意义(P<0.01);GDM组危险等位基因G分布频率(64.6%)明显高于对照组(47.0%)(P<0.01);GDM组GG、GA、AA基因亚型空腹血糖水平分别为(6.30(6.00,6.70)、5.90(5.55,6.20)、5.70(5.18,6.10)mmol/L),胰岛素抵抗指数分别为(4.04±0.35、3.72±0.47、3.43±0.10),除GG、GA亚型空腹血糖水平差异无统计学意义(P>0.05),3个亚型间空腹血糖水平和胰岛素抵抗指数比较差异均有统计学意义(P<0.05)。结论 UTS-Ⅱ基因rs228651位点(G/A)多态性与GDM易感性有关。
Objective To investigate the association between the polymorphism of rs228651 gene of urotensin-2 (UTS-Ⅱ) gene and genetic susceptibility to gestational diabetes mellitus (GDM). Methods 120 patients with GDM (GDM group) and 100 normal pregnant women with impaired glucose tolerance (control group) were genotyped by restriction fragment length polymorphism (PCR) to detect the rs228651 locus in UTS-Ⅱ gene. Relationship between rs228651 Polymorphism and Genetic Susceptibility to. Results The fasting blood glucose, fasting insulin, total cholesterol, insulin resistance index and β cell function index of GDM group were (6.05 ± 0.51) mmol / L, 14.20 (13.22,15.60) mu / L and 5.85 , 3.82 ± 0.47 and 116.55 (94.19, 135.79) in control group and 4.40 ± 0.40 mmol / L and 10.19 (9.63,10.70) mu / L and 4.70 (4.50 and 4.90) mmol / L and 2.90 ± 0.24 and 225.56 (174.48,314.64). There was significant difference between the two groups (P <0.05). The frequency of GG and AA subtypes in URS-Ⅱ gene rs228651 in GDM group was 44.2% and 15.0% , 29.0% respectively) (P <0.01). The distribution frequency of G allele in GDM group (64.6%) was significantly higher than that in control group (47.0%) (P <0.01) The fasting blood glucose levels of AA genotypes were (6.30 (6.00, 6.70), 5.90 (5.55,6.20), 5.70 (5.18,6.10) mmol / L respectively) and the insulin resistance index were (4.04 ± 0.35,3.72 ± 0.47,3.43 ± 0.10). There was no significant difference in fasting plasma glucose between GG and GA subtypes (P> 0.05). There was significant difference in fasting blood glucose and insulin resistance between the three subtypes (P <0.05). Conclusion UTS-Ⅱ gene rs228651 (G / A) polymorphism is associated with GDM susceptibility.
