目的:研究栀子柏皮汤对阳黄证模型大鼠的治疗作用与作用机制。方法:采用以异硫氰酸苯酯(APIT)ig诱导大鼠肝损伤基础上,同时结合高温高湿的环境因素、高糖高脂的饮食因素的方法,复制阳黄证大鼠模型,施以栀子柏皮汤低、中、高剂量(2.0,4.0,8.0 g·kg-1)治疗,观察大鼠的一般生物学状态,检测血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、总胆红素(TBIL)、胆汁酸(TBA)和补体C3,C4水平,以及肝组织中总超氧化物歧化酶(T-SOD)活性和脂质过氧化产物丙二醛(MDA)含量,光镜下观察大鼠肝组织病理变化。结果:与正常组比较,模型组大鼠血清中ALT,AST,TBIL,TBA含量均有极显著升高,肝组织中MDA的含量极显著增加、SOD的活性极显著降低(均P<0.01)。与阳黄证模型组相比,栀子柏皮汤中、高剂量组均能显著降低大鼠血清中ALT,AST,TBIL,TBA和补体C3,C4表达水平(均P<0.01),显著提高肝SOD活性(P<0.05,P<0.01),且栀子柏皮汤中剂量组可以显著降低肝MDA含量(P<0.05),病理切片显示二者均可不同程度地减轻肝组织损伤。结论:栀子柏皮汤对阳黄证模型大鼠有治疗作用,清除自由基、抑制炎症反应从而减轻肝脏损伤,可能为其退黄机制。
Objective: To study the therapeutic effect and mechanism of Gardenia Bibi Decoction on yang-Huang syndrome model rats. Methods: Rat liver injury was induced by phenylisothiocyanate (APIT) ig, and combined with environmental factors of high temperature and humidity, diet factors of high glucose and high fat, The rats were treated with 2.0, 4.0 and 8.0 g · kg-1 of low, medium and high doses of Gardenia Bibi Tang for a few days. The serum levels of alanine aminotransferase (ALT), aspartate (AST), total bilirubin (TBIL), bile acid (TBA), complement C3 and C4, as well as total superoxide dismutase (T-SOD) activity and lipid peroxidation The content of malondialdehyde (MDA) in rat liver tissue was observed under light microscope. Results: Compared with the normal group, the levels of ALT, AST, TBIL and TBA in the model group were significantly increased, the content of MDA in the liver tissue increased significantly and the activity of SOD was significantly decreased (all P <0.01) . Compared with the Yang-Huang syndrome model group, both the medium-dose and high-dose Zhizhibibai Tang groups could significantly reduce the levels of ALT, AST, TBIL, TBA, complement C3 and C4 in serum (all P <0.01) Liver SOD activity (P <0.05, P <0.01), and medium dose group of Zhizhipubibai can significantly reduce the content of MDA in the liver (P <0.05). Pathological examination showed that both of them could alleviate the liver injury in varying degrees. Conclusion: Gardenia Bibi Tang has the therapeutic effect on rats with yang-Huang syndrome, which can eliminate the free radicals, inhibit the inflammatory reaction and relieve the liver damage.
