目的采用反转录聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)检测咬干扰对实验大鼠颞下颌关节软骨的炎性因子表达。方法选用6周龄雌性SD大鼠9只,随机分为咬紊乱组、去除咬紊乱组和正常对照组,每组3只。制造咬干扰动物模型。6周后,咬紊乱组不做处理,去除咬紊乱组去除咬干扰,正常对照组不做处理。8周后切取大鼠右侧颞下颌关节,采用RT-PCR方法检测颞下颌关节软骨中白细胞介素1(interleukin-1,IL-1),白细胞介素6(interleukin-6,IL-6),肿瘤坏死因子α(tumornecrosis factorα,TNF-α)及γ干扰素(interferon-γ,IFN-γ)的表达。结果在咬干扰条件下,RT-PCR检测显示炎性因子IL-1、IL-6及TNF-α在各组大鼠中均有表达,但咬紊乱组和去除咬紊乱组的炎性因子表达均高于对照组,咬紊乱组IL-6和TNF-α的表达高于去除咬紊乱组;3组大鼠均未检测到IFN-γ的表达。结论咬干扰条件下,炎性因子IL-1、IL-6及TNF-α在颞下颌关节软骨中的表达增强,提示炎性因子与实验大鼠髁突软骨病理性改建活动有相关性。
Objective To detect the expression of inflammatory cytokines in the temporomandibular joint cartilage of experimental rats by reverse transcription polymerase chain reaction (RT-PCR). Methods Nine 6-week-old female Sprague-Dawley rats were randomly divided into 2 groups: bite disheartened group and normal control group, with 3 in each group. Create bite interference animal model. After 6 weeks, the bites were disorganized, the bites were removed and the bites were removed. The normal control group was not treated. After 8 weeks, the temporomandibular joint on the right side of the rat was cut out, and the expression of interleukin-1 (IL-1), interleukin-6 (IL-6) , Tumor necrosis factor α (TNF-α) and interferon-γ (IFN-γ) were detected by ELISA. Results The results of RT-PCR showed that the inflammatory factors IL-1, IL-6 and TNF-α were expressed in all the rats in the bite group, but the bite group and the biliary group The expression of IL-6 and TNF-α was higher in the bites of cholestasis group than in the control group. The expression of IFN-γ was not detected in all three groups. Conclusion The expression of inflammatory cytokines IL-1, IL-6 and TNF-α in temporomandibular joint cartilage were increased under bite disturbance, suggesting that inflammatory cytokines correlate with condylar cartilage pathological remodeling in experimental rats.
