目的:优选大孔树脂纯化展毛地椒总黄酮的工艺参数。方法:采用紫外分光光度法测定总黄酮含量,以总黄酮的吸附率和解吸率为指标,考察大孔树脂型号、上样液浓度、上样流速、洗脱剂种类及其用量等影响因素,优选最佳分离纯化条件。结果:AB-8树脂具有较好的吸附率和解吸率,最佳纯化工艺:上样液浓度为0.2 g/m L,上样流速为2 BV/h,上样体积为2 BV,洗脱溶剂为50%乙醇,洗脱体积为5 BV,径高比为1/6,富集物中展毛地椒总黄酮含量达到26.32%。结论:AB-8型大孔树脂能较好地用于分离纯化展毛地椒总黄酮。
