目的对不同产地、不同生长年限人参进行淀粉酶(AMY)、酯酶(Est)和酸性磷酸酯酶(ACP)活力比较。方法用中性缓冲溶液提取总蛋白,应用3,5-二硝基水杨酸比色法测定AMY活力;乙酸-α-萘酯比色法测定Est活力;对硝基酚比色法测定ACP活力。结果不同产地人参AMY、Est、ACP活力均存在很大差异。取不同产地、相同生长年限酶活力平均值进行不同年限的酶活力比较,可知4年与5年生人参3种水解酶活力均无明显差异。结论 AMY、Est、ACP活力可以作为评价人参质量的内在指标之一。
Objective To compare the activities of amylase (AMY), esterase (Est) and acid phosphatase (ACP) in ginseng from different habitats and growth stages. Methods The total protein was extracted by neutral buffer solution. The activity of AMY was measured by 3,5-dinitrosalicylic acid colorimetric method. The activity of Est was measured by acetic acid-α-naphthyl ester colorimetry. The activity of ACP vitality. Results There were great differences in the activities of AMY, Est and ACP in different areas. Take different origin, the same growth of the average enzyme activity of different years of enzyme activity comparison, we can see 4-year and 5-year-old ginseng three kinds of hydrolase activity were no significant difference. Conclusion AMY, Est, ACP activity can be used as one of the intrinsic indicators to evaluate the quality of ginseng.
