目的:采用高效液相色谱法测定不同产地野生滇龙胆中龙胆苦苷、獐牙菜苦苷及獐牙菜苷的含量。方法:岛津Shim-pack VP-ODS柱(4.6 mm×150 mm,5μm),流动相甲醇-水(30∶70),流速1.0 mL·min-1,龙胆苦苷检测波长270 nm,獐牙菜苦苷及獐牙菜苷检测波长245 nm,柱温25℃。结果:龙胆苦苷、獐牙菜苦苷、獐牙菜苷的线性范围为0.4~2 mg·L-1;相关系数分别为0.999 7,0.999 6,0.998 7;回收率分别为100.04%(RSD 1.49%),98.48%(RSD 1.67%),98.46%(RSD 1.15%)结论:用本法测定不同产地野生滇龙胆中3种成分含量,方法可行,结果可靠,为野生滇龙胆资源的开发与利用提供资料。
OBJECTIVE: To determine the contents of gentiopicroside, swertiarin and swertia glycoside in Gentiana yunnanensis Gentiana in different habitats by high performance liquid chromatography (HPLC). Methods Shimadzu Shim-pack VP-ODS column (4.6 mm × 150 mm, 5 μm), mobile phase methanol-water (30:70), flow rate 1.0 mL · min-1, gentiopicroside detection wavelength 270 nm, Carnosine and swertia glycosides detection wavelength 245 nm, column temperature 25 ℃. Results: The linear ranges of gentiopicroside, swertiarin and swertiadenin were 0.4-2 mg · L-1, the correlation coefficients were 0.999 7, 0.999 6 and 0.998 7, respectively. The recovery rates were 100.04% RSD 1.49%), 98.48% (RSD 1.67%) and 98.46% (RSD 1.15%) respectively. Conclusion: The method can be used to determine the content of three components in Gentiana yunnanensis from different habitats. The method is reliable and the results are reliable.
