目的:观察益气化瘀口服液治疗N-甲基-N-亚硝脲(MNU)诱导大鼠视网膜变性的疗效。方法:雌性SD大鼠150只随机分为5组。益气化瘀口服液低、中、高剂量组分别给予益气化瘀口服液1.5、3.0、6.0 mL/d灌胃,正常对照组和模型组以等体积蒸馏水灌胃,3次/d,连续7 d。给药7 d后,益气化瘀口服液组和模型组大鼠予MNU40 mg/kg,ip;正常对照组注射等体积生理盐水。观察MNU处理后1、3、7 d各组大鼠视网膜电图(ERG)a波及b波的变化,并取眼球进行光学显微镜和视网膜透视电子显微镜的组织学观察。结果:正常对照组大鼠不同时间点ERG a波及b波的振幅差异无统计学意义(P>0.05);模型组在MNU处理后ERG a波及b波的振幅均呈持续性、进行性下降,益气化瘀口服液可明显抑制ERG a波及b波振幅的降低(P<0.05)。眼病理结果,与模型组比较,益气化瘀口服液组大鼠视网膜光感受器细胞损伤明显减轻。结论:益气化瘀口服液能减轻MNU对大鼠视网膜的损伤程度,促进MNU诱导的大鼠视网膜光感受器细胞损伤的功能恢复。
Objective: To observe the curative effect of Yiqi Huayu oral liquid on retinal degeneration induced by N-methyl-N-nitrosourea (MNU) in rats. Methods: 150 female SD rats were randomly divided into 5 groups. Yiqi Huayu oral solution of low, medium and high dose groups were given Yiqi Huayu oral solution 1.5,3.0,6.0 mL / d gavage, normal control group and model group with equal volume of distilled water, 3 times / d, For 7 days. After 7 days of administration, the rats in Yiqi Huayu oral liquid group and model group were given MNU 40 mg / kg, ip; normal control group were injected with equal volume of normal saline. The change of a wave and b wave of electroretinogram (ERG) of rats in each group was observed on the 1st, 3rd, 7th day after MNU treatment. The eyeballs were observed for histological observation by light microscope and transmission electron microscopy. Results: The amplitudes of ERG a and b wave in normal control group were not significantly different at different time points (P> 0.05). The amplitudes of ERG a and b wave in MNU group were both persistent and progressive, Yiqi Huayu oral liquid can significantly inhibit ERG a and b wave amplitude decreased (P <0.05). Eye pathology results, compared with the model group, Yiqi Huayu oral liquid rat retinal photoreceptor cell damage was significantly reduced. Conclusion: Yiqi Huayu oral liquid can reduce the damage of MNU to the retina of rats and promote the functional recovery of MNU-induced retinal photoreceptor cell injury in rats.
