目的:建立反相高效液相色谱法同时测定路边青药材中咖啡酸、槲皮素-3-O-葡萄糖苷、山柰酚-3-O-葡萄糖苷、山柰酚含量的方法。方法:采用Agilent C_(18)色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-0.5%甲酸水溶液梯度洗脱,流速0.8 m L·min~(-1),柱温25℃,检测波长340 nm。结果:咖啡酸、槲皮素-3-O-葡萄糖苷、山柰酚-3-O-葡萄糖苷、山柰酚分别在0.045~1.130,0.095~2.370,0.105~2.635,0.041~1.030μg线性关系良好,平均加样回收率(n=6)分别为99.0%,98.5%,97.7%,100.7%,RSD分别为1.0%,0.9%,1.0%,1.6%。结论:该方法简便、快捷,重复性好,可用于路边青药材的质量控制。
Objective: To establish a method for the simultaneous determination of caffeic acid, quercetin-3-O-glucoside, kaempferol-3-O-glucoside and kaempferol in Curcuma longanside by reversed-phase high performance liquid chromatography. METHODS: The mobile phase was eluted with a mobile phase of acetonitrile-0.5% formic acid in a gradient of 0.8 mL · L -1 on an Agilent C 18 column (4.6 mm × 250 mm, 5 μm) Wavelength 340 nm. Results: The linear relationships of caffeic acid, quercetin -3-O-glucoside, kaempferol-3-O-glucoside and kaempferol were 0.045 ~ 1.130,0.095 ~ 2.370,0.105 ~ 2.635,0.041 ~ 1.030μg The average recoveries (n = 6) were 99.0%, 98.5%, 97.7% and 100.7%, respectively. The RSDs were 1.0%, 0.9%, 1.0% and 1.6% respectively. Conclusion: The method is simple, rapid, reproducible and can be used for the quality control of roadside green medicinal materials.
