目的探讨强直性脊柱炎患者外周血T细胞凋亡相关蛋白的表达以及T细胞凋亡状况,分析其在疾病免疫发病机制的作用。方法CT检查患者骶髂关节并分级。尼龙棉柱法分离患者外周血T细胞,PI染色结合流式细胞术检测T细胞凋亡率,免疫组化法检测T细胞促凋亡蛋白FADD、Caspase-3、Caspase-8和抗凋亡蛋白FLIP表达。结果与健康对照相比,强直性脊柱炎患者外周血T细胞凋亡率明显降低(P<0.05);促凋亡蛋白FADD、Caspase-3、Caspase-8百分率均明显降低(P<0.05),抗凋亡蛋白FLIP百分率明显升高(P<0.05)。结论强直性脊柱炎患者外周血T细胞凋亡蛋白及抗凋亡蛋白表达异常和T细胞凋亡不足与患者免疫学发病机制相关。
Objective To investigate the expression of apoptosis-related protein and the apoptosis of T cells in peripheral blood of patients with ankylosing spondylitis, and to analyze its role in the pathogenesis of immune diseases. Methods CT examination of patients with sacroiliac joint and grading. Peripheral blood T cells were isolated by nylon cotton column method. T cell apoptosis rate was detected by PI staining and flow cytometry. The expressions of FADD, Caspase-3, Caspase-8 and anti-apoptotic protein were detected by immunohistochemistry FLIP expression. Results Compared with healthy control, the apoptosis rate of T cells in peripheral blood of patients with ankylosing spondylitis was significantly lower (P <0.05), the percentage of FADD, Caspase-3 and Caspase-8 were significantly decreased (P <0.05) The percentage of anti-apoptotic protein FLIP was significantly increased (P <0.05). Conclusion The abnormal expression of anti-apoptotic protein and anti-apoptotic protein in peripheral blood of patients with ankylosing spondylitis is related to the immunological pathogenesis of patients with ankylosing spondylitis.
