目的研究槲皮素对敌敌畏诱导的大鼠肺毒性的影响。方法将80只雄性Wistar大鼠按体重随机分为8组:正常对照组,槲皮素低剂量组,槲皮素中剂量组,槲皮素高剂量组,敌敌畏处理组,低剂量槲皮素与敌敌畏联合组,中剂量槲皮素与敌敌畏联合组,高剂量槲皮素与敌敌畏联合组。槲皮素经灌胃给药,敌敌畏通过饮水给予大鼠,连续给药90 d。检测肺脏组织中超氧化物歧化酶(SOD),过氧化氢酶(CAT),谷胱甘肽过氧化物酶(GPx)的活性及丙二醛含量(MDA),并观察肺脏组织病理学变化。结果与对照组大鼠相比,敌敌畏组大鼠肺脏组织丙二醛(MDA)含量及抗氧化物酶类活性显著升高(P<0.01)。当大鼠同时给予敌敌畏和槲皮素后,上述指标均有下降趋势。与敌敌畏组相比,敌敌畏与高剂量槲皮素组大鼠肺脏组织MDA含量显著下降,且超氧化物歧化酶(SOD)、过氧化物酶(CAT)和谷胱甘肽过氧化物酶(GPx)活性显著降低(P<0.05)。上述指标在3个剂量的单独槲皮素组与对照组相比没有统计学差异(P>0.05)。肺脏病理组织学检测结果显示,敌敌畏组大鼠肺脏组织可见肺泡间质增厚、单核细胞浸润、肺气肿;这种病理学改变在高剂量槲皮素与敌敌畏联合组有所改善。结论槲皮素对敌敌畏所诱导的肺毒性具有部分保护作用。
Objective To study the effect of quercetin on the toxicity of dichlorvos in rats. Methods Eighty male Wistar rats were randomly divided into 8 groups according to body weight: normal control group, quercetin low dose group, quercetin middle dose group, quercetin high dose group, dichlorvos treatment group, low dose quercetin In combination with dichlorvos, the combination of quercetin with dichlorvos and the combination of high dose quercetin and dichlorvos. Quercetin was intragastrically administered, and dichlorvos were given to rats by drinking water for 90 days. The activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) and the content of malondialdehyde (MDA) in lung tissue were detected. The pathological changes of lung tissue were observed. Results Compared with the control group, the content of malondialdehyde (MDA) and the activity of antioxidant enzymes in the lung tissue of the dichlorvos group were significantly increased (P <0.01). When the rats given both dichlorvos and quercetin, the above indicators have a downward trend. Compared with the dichlorvos group, the content of MDA in the lung tissue of the rats in the dichlorvos and high dose quercetin groups decreased significantly, and the contents of superoxide dismutase (SOD), peroxidase (CAT) and glutathione peroxidase GPx) activity was significantly lower (P <0.05). The above index in the three doses of quercetin alone compared with the control group was no significant difference (P> 0.05). Histopathological examination of lung showed that alveolar septa thickened, mononuclear cell infiltrate and emphysema were found in the lung tissue of the dichlorvos group rats. The pathological changes were improved in the combination of high dose quercetin and dichlorvos. Conclusion Quercetin has some protective effects on lung toxicity induced by dichlorvos.
