目的探究丙烯酰胺(ACR)染毒对孕哺期大鼠胼胝体髓鞘碱性蛋白(MBP)和髓鞘蛋白脂蛋白(MPLP)表达的影响。方法 32只SPF级SD大鼠随机分为对照组,4.5、9及18 mg/kg ACR染毒组,在大鼠妊娠第15天至分娩后第13天灌胃染毒。观测大鼠体重和步态的变化,通过免疫组化方法检测大鼠胼胝体MBP和MPLP的表达。结果随着ACR染毒剂量的增加,大鼠体重降低,呈剂量依赖性。与对照组和4.5 mg/kg ACR组相比较,9和18 mg/kg ACR组步态评分显著增加(P<0.01);胼胝体MBP和MPLP表达显著减少(P<0.05)。与对照组相比较,9 mg/kg组大鼠大脑MPLP明显减少(P<0.05),而18 mg/kg组MPLP和MBP的表达均显著减少(P<0.01)。结论 ACR染毒可能会通过减少孕哺期大鼠胼胝体MBP和MPLP的表达,损害其髓鞘的结构和功能,影响中枢神经系统信息的传导功能。
Objective To investigate the effect of acrylamide (ACR) on the expression of myelin basic protein (MBP) and myelin lipoprotein (MPLP) in corpus callosum of rats during pregnancy and lactation. Methods Thirty-two Sprague-Dawley (SD) SD rats were randomly divided into control, 4.5, 9 and 18 mg / kg ACR groups. The rats were intragastrically administered on day 15 of gestation until 13 days after delivery. The changes of body weight and gait were observed. The expression of MBP and MPLP in the corpus callosum was detected by immunohistochemistry. Results With the increase of ACR dose, rats lost weight in a dose-dependent manner. Compared with the control group and 4.5 mg / kg ACR group, the gait scores of 9 and 18 mg / kg ACR group were significantly increased (P <0.01). The expression of MBP and MPLP in the corpus callosum was significantly decreased (P <0.05). Compared with the control group, MPLP in 9 mg / kg group decreased significantly (P <0.05), while MPLP and MBP decreased in 18 mg / kg group (P <0.01). Conclusion ACR could reduce the expression of MBP and MPLP in the corpus callosum during pregnancy and lactation, impair the structure and function of myelin sheath and affect the conduction of central nervous system information.
