应用3H-胸腺嘧啶核苷(3H-TdR)掺入法和四甲基偶氮唑MTT比色法,探讨了人重组白细胞介素1(interleukin-1,IL-1)和白细胞介素6(interleukin-6,IL-6)对成人肾小球系膜细胞增殖的影响。结果发现,在无血清培养条件下,γIL-1(recombinantIL-1)和γIL-6(recombinantIL-6)均不能刺激系膜细胞的繁殖;而在有少量胎牛血清存在的条件下,γIL-6可以明显地促进系膜细胞增殖,但γIL-1无此作用。我们的实验还表明,γIL-1和γIL-6均能促进系膜细胞合成PAF。IL-1和IL-6的这些作用提示,它们在肾小球免疫病理损伤过程中可能发挥重要作用。GroupB表示培养液中含低浓度血清(5%FCS)表2IL-1对系膜细胞增殖的影响(MTT法)GroupA、GroupB意义同表13H-TdR掺入法及MTT法的结果均显示,无论培养基中有无血清存在,γIL-1在一个较大的浓度范围内(10~100Ou/well),都不能促进系膜细胞增殖(与对照组相比P>0.05)。2.2IL-6对系膜细胞增殖的影响研究发现,在无血清培养条件下,γIL-6并不表现促细胞生长的作用?
The effects of human recombinant interleukin-1 (IL-1) and interleukin-6 (IL-1) interleukin-6, IL-6) on the proliferation of adult glomerular mesangial cells. The results showed that, γIL-1 (recombinant IL-1) and recombinant IL-6 could not stimulate the proliferation of mesangial cells in serum-free culture conditions; while in the presence of a small amount of fetal bovine serum, 6 can obviously promote mesangial cell proliferation, but γIL-1 does not have this effect.
