目的 探索用维甲酸(RA)联合干扰素-γ(IFN-γ)协同诱导分化治疗脑胶质瘤的新方案。方法应用改良MTT法,系统分析RA/IFN-γ协同调控脑胶质瘤细胞增殖的量效、时效关系。结果 RA及IFN--γ单独应用时,在药理可达浓度(分别为10~(-6)M及40 IU/ml)水平对体外培养的脑胶质瘤细胞具有一定的抑制增殖效应,但作用较弱;RA(特别是9-cisRA)与IFN-γ联用时显示出较强的协同抗增殖作用,诱导分化效应大大增强。结论 RA/IFN-γ协同诱导分化治疗能有效逆转脑胶质瘤细胞的恶性增殖表型。
OBJECTIVE: To explore a novel approach for the treatment of glioma by the synergistic induction and differentiation of retinoic acid (RA) combined with interferon-γ (IFN-γ). Methods The improved MTT method was used to systematically analyze the dose-response and time-effect relationship of RA / IFN-γ synergistically regulating glioma cell proliferation. Results When RA and IFN - γ were administered alone, the proliferation of glioma cells cultured in vitro was inhibited to some extent at pharmacologically acceptable concentrations (10 ~ (-6) M and 40 IU / ml, respectively) The combination of RA (especially 9-cisRA) and IFN-γ showed a strong synergistic antiproliferative effect, and the differentiation effect was greatly enhanced. Conclusion The synergistic induction and differentiation of RA / IFN-γ can effectively reverse the malignant phenotype of glioma cells.
