L-精氨酸对宫内发育迟缓大鼠肾功能的影响及机制探讨

目的研究哺乳期用L-精氨酸(L-Arg)对宫内发育迟缓(IUGR)大鼠肾功能的影响及机制探讨。方法给予孕鼠10%低蛋白饲料制造IUGR仔鼠模型,并在哺乳期(21d内)给母鼠含21%蛋白饲料,添加L-Arg干预者为干预组(LT组),不添加者为IUGR组;正常对照组(NC组)全程予含21%蛋白饲料喂养。于生后1 w、3 w、2月和3月龄,留取血清标本测定肌酐(Cr)、尿素氮(BUN)和NO浓度;取后3个时间点,留取尿液,测尿肌酐(UCr)和24h尿蛋白定量。结果(1)与NC组比较,IUGR组在4个时间点血清NO的浓度均有显著降低;而LT组与正常组比较无显著差异;LT组与IUGR组相比,3 w、2月和3月龄血清NO的浓度均明显增加。(2)3w和2月龄时,三组仔鼠的24h尿蛋白定量比较无显著差异;3月龄时,IUGR组显著高于NC组和LT组。(3)三组仔鼠在4个观察点比较Cr无显著差异;2月龄时IUGR组与NC组比较,UCr有显著差异;3月龄时与NC组和LT组比较,IUGR组大鼠UCr差异有显著性。(4)与NC组和LT组比较,IUGR组CCr水平在3w、2月和3月龄均有显著减低。结论生后早期给予L-Arg干预,IUGR大鼠血清NO浓度和CCr提高、24h尿蛋白减低;IUGR仔鼠体内NO降低是CCr降低和24h蛋白尿增加的重要原因之一。 Objective To investigate the effect of L-arginine (L-Arg) on ​​renal function in rats with intrauterine growth retardation (IUGR) during lactation. Methods IUGR pups model was induced by 10% low-protein diet for pregnant rats, and 21% protein diet was given to female rats during lactation (21d). Intervention group (L-Arg) IUGR group; normal control group (NC group) to 21% protein feed throughout the feeding. Serum samples were collected for serum creatinine (Cr), blood urea nitrogen (BUN) and NO concentrations at 1 w, 3 w, 2 months and 3 months postnatal days. (UCr) and 24 h urinary protein. Results (1) Compared with NC group, NO concentration in serum of IUGR group was significantly decreased at 4 time points, but there was no significant difference between LT group and normal control group. Compared with IUGR group, 3-month serum NO concentrations were significantly increased. (2) There was no significant difference in 24 h urinary protein among three groups of offspring at 3w and 2 months of age; IUGR group was significantly higher than NC group and LT group at 3 months. (3) There was no significant difference in Cr between the four groups at 4 observation points; UCr was significantly different between IUGR group and NC group at 2 months of age; compared with NC group and LT group at 3 months, IUGR rats UCr difference was significant. (4) Compared with NC group and LT group, CCr levels in IUGR group were significantly decreased at 3w, 2 and 3 months. Conclusion L-Arg intervention in early postnatal period, IUGR rat serum NO concentration and CCr increased 24h urinary protein decreased; IUGR offspring NO reduction in rats is one of the important reasons for the decreased CCr and 24h proteinuria.
