目的:研究D-半乳糖诱导的衰老大鼠模型在行为学上发生的变化,对其进行阶段性的Morris水迷宫测试,分析D-半乳糖致衰老大鼠在不同时期的空间学习记忆能力的改变,从而论证D-半乳糖在造脑衰老模型方面的可行性。方法:5% D-半乳糖100 mg/(kg·d)连续腹腔注射大鼠9周,应用Morris水迷宫方法分阶段进行行为学测试。结果:D-半乳糖给药组第4周和第7周的逃避潜伏期、游泳速度及穿越平台区次数与空白组比较差异不显著(P>0.05),造模第8周和第9周逃避潜伏期、游泳速度及穿越平台区次数与空白组比较差异显著(P<0.05),但第8周与第9周的模型组之间没有显著性差异(P>0.05)。结论:采用Morris水迷宫法检测D-半乳糖致衰老模型是否成功具有较强的可行性,且把造模周期定为8周比较恰当。
OBJECTIVE: To study the behavioral changes in the aging rat model induced by D-galactose, and to conduct a phased Morris water maze test to analyze the spatial learning and memory ability of D-galactose-induced aging rats at different stages Change, thus demonstrating the feasibility of D-galactose in the model of brain aging. Methods: Rats were injected intraperitoneally with 5% D-galactose 100 mg / (kg · d) for 9 weeks. Morris water maze method was used to conduct behavioral tests in stages. Results: The escape latency, swimming speed and the number of passing through the platform in D-galactose group were not significantly different from those in blank group at the 4th and 7th week (P> 0.05) The latency, swimming speed and the number of crossing the platform area were significantly different from those in the blank group (P <0.05). However, there was no significant difference between the model group at the 8th week and the 9th week (P> 0.05). Conclusion: It is feasible to use Morris water maze test to detect the success of D-galactose-induced aging model. It is more appropriate to set the modeling cycle as 8 weeks.
