目的观察益气化瘀解毒方对脂多糖(lipopolysaccharide,LPS)所致大鼠急性呼吸窘迫综合征(acute respiratory distress syndrome,ARDS)的保护作用并探讨其作用机制。方法 30只SD大鼠随机分为空白组、模型组及益气化瘀解毒方低、中、高剂量组5组,每组6只。尾静脉注射LPS建立ARDS模型。治疗组益气化瘀解毒方灌胃,造模后16小时处死大鼠收集标本,测定大鼠肺系数、肺通透指数、核转录因子-κB p50、IκBα、p38的蛋白表达及光镜下观察肺组织病理学改变。结果益气化瘀解毒方减少肺损伤大鼠肺泡结构破坏、肺水肿及炎性细胞浸润,降低肺系数(P<0.05)与肺通透指数(P<0.01),并在一定程度减少核转录因子-κB p50、丝裂原活化蛋白激酶p38,增加抑制因子IκBα的蛋白表达。结论益气化瘀解毒方对LPS诱导的ARDS有一定的保护作用,其机制与抑制NF-κB/p38MAPK炎性反应信号通路有关。
Objective To observe the protective effect of Yiqi Huayu Jiedu Decoction on acute respiratory distress syndrome (ARDS) induced by lipopolysaccharide (LPS) in rats and its mechanism. Methods Thirty SD rats were randomly divided into blank group, model group and Yiqi Huayu Jiedu Fang low, medium and high dose group of 5 groups, 6 in each group. ARDS models were established by tail vein injection of LPS. The treatment group was given Yiqi Huayu Jiedu Decoction and the rats were sacrificed 16 hours after model establishment. The lung index, pulmonary permeability index, nuclear factor-κB p50, IκBα, Lung histopathology was observed. Results Yiqi Huayu Jiedu Decoction could reduce alveolar structural damage, pulmonary edema and inflammatory cell infiltration, reduce pulmonary coefficient (P <0.05) and pulmonary permeability index (P <0.01), and to a certain extent reduce nuclear translocation Factor-κB p50, mitogen-activated protein kinase p38, increases the protein expression of the inhibitory factor IκBα. Conclusion Yiqi Huayu Jiedu Decoction has some protective effect on LPS-induced ARDS, and its mechanism is related to the inhibition of NF-κB / p38MAPK inflammatory response signal pathway.
