目的探讨亚低温对脑出血后内源性神经干细胞(neural stem cells,NSCs)增殖及迁移的影响。方法采用胶原酶脑内立体定向注射制作脑出血模型。模型成功后将大鼠随机分为对照组和亚低温组,每组分为7、14、21、28 d亚组。应用腹腔注射溴脱氧尿嘧啶核苷(Bromodeoxyuridine,BrdU)标记增殖的NSCs。采用免疫组织化学方法动态观察同侧侧脑室下区(subventricular zone,SVZ)及血肿周围BrdU阳性细胞的变化。结果对照组同侧SVZ及血肿周围BrdU阳性细胞在出血后7 d时开始增加,14 d达到高峰,21 d后开始下降。亚低温组显著增加各时间点SVZ及血肿周围BrdU阳性细胞(P<0.01)。结论亚低温能促进脑出血后内源性NSCs增殖及迁移。
Objective To investigate the effects of mild hypothermia on the proliferation and migration of endogenous neural stem cells (NSCs) after intracerebral hemorrhage. Methods Intracerebral stereotactic injection of collagenase was used to make intracerebral hemorrhage model. After successful model, the rats were randomly divided into control group and mild hypothermia group, each group was divided into 7, 14, 21, 28 d subgroups. Proliferation of NSCs was marked by intraperitoneal injection of Bromodeoxyuridine (BrdU). The changes of ipsilateral subventricular zone (SVZ) and BrdU positive cells around the hematoma were observed by immunohistochemistry. Results The ipsilateral SVZ and the BrdU positive cells around the hematoma began to increase at 7 days after hemorrhage in the control group, reached the peak on the 14th day and began to decrease after 21 days. The mild hypothermia group significantly increased SVZ and BrdU positive cells around the hematoma at each time point (P <0.01). Conclusion Mild hypothermia can promote the proliferation and migration of endogenous NSCs after intracerebral hemorrhage.
