目的:探讨丙丁酚(普罗布考)对氧化低密度脂蛋白(oxLDL)诱导人脐静脉血管内皮细胞(HUVEC)损伤的保护作用,以及对NF-κB和IκB-α表达的调节作用。方法:MTT法分别检测oxLDL不同浓度(1g/ml、10g/ml、100g/ml)处理HUVEC不同时间(12h、24h、48h)细胞增殖情况;5g/mL丙丁酚预处理HUVEC24小时,MTT检测细胞增殖情况;Western blot检测oxLDL处理细胞及丙丁酚预处理后NF-κB和IκB-α蛋白表达水平。结果:oxLDL处理HUVEC细胞24小时后,对细胞增殖具有抑制作用,Western blot结果显示NF-κB表达显著上调,IκB-α蛋白表达下调。而用5ug/mL丙丁酚预处理可以明显缓解细胞增殖抑制,NF-κB和IκB-α蛋白的表达也恢复至正常水平。结论:丙丁酚可显著缓解oxLDL诱导的HUVEC细胞增殖抑制,并且NF-κB和IκB-α表达的改变可能参与此过程。
OBJECTIVE: To investigate the protective effect of probucol on oxidative low density lipoprotein (oxLDL) -induced injury of human umbilical vein endothelial cells (HUVECs) and the regulation of NF-κB and IκB-α expression. Methods: The proliferation of HUVECs treated with oxLDL at different concentrations (1g / ml, 10g / ml, 100g / ml) for different time (12h, 24h, 48h) were detected by MTT assay. HUVECs were pretreated with 5g / Western blot was used to detect the expression of NF-κB and IκB-α in oxLDL-treated cells and probucol preconditioned cells. Results: OxLDL treatment of HUVEC cells 24 hours after inhibition of cell proliferation, Western blot results showed that NF-κB expression was significantly increased, IκB-α protein expression down. Pretreatment with 5ug / mL probucol could obviously inhibit the cell proliferation and the expression of NF-κB and IκB-α also returned to normal levels. CONCLUSION: Probucol can relieve the oxLDL-induced inhibition of HUVEC proliferation. The changes of NF-κB and IκB-α may be involved in this process.
