目的:探讨异甘草酸镁注射液对大鼠梗阻性黄疸肝损伤的保护作用。方法:选用健康雄性SD大鼠32只,随机分成4组,每组各8只,分别为胆总管结扎+异甘草酸镁注射液常规剂量组(BMc组);胆总管结扎+异甘草酸镁注射液高剂量组(BMh组);胆总管结扎+0.9%生理盐水注射液组(BN组)和假手术组(Sham组)。BN和Sham组以生理盐水30 mg/kg/日腹腔注射,BMc组以异甘草酸镁30 mg/kg/日腹腔注射,BMh组以异甘草酸镁60 mg/kg/日腹腔注射。术后不同时间对各组大鼠眼眶取血并获得血清。在全自动生化分析仪器检测肝功能指标:丙氨酸氨基转移酶(Alanine aminotransferase,ALT)、天冬氨酸氨基转移酶(Aspartate aminotransferase,AST)、直接胆红素(Direct bilirubin,DBil)。结果:BN、BMc和BMh组大鼠在胆管结扎后血清中ALT和AST指标明显升高。随着梗阻时间的增加,BM和BMh组大鼠血清中ALT和AST的水平均明显低于BN组(P<0.01),但两组之间并无明显区别。手术后BN、BMc和BMh组大鼠在胆管结扎后血清中D-Bil水平明显高于Sham组(P<0.01),但随着梗阻时间的延长,各组间D-Bil的水平变化不明显。结论:异甘草酸镁注射液可以有效降低梗阻性黄疸大鼠血清转氨酶水平,对肝功能有明显的保护作用。
Objective: To investigate the protective effect of magnesium isrimazole injection on hepatic injury induced by obstructive jaundice in rats. Methods: Thirty-two healthy male Sprague-Dawley rats were randomly divided into 4 groups with 8 rats in each group. The rats were randomly divided into common bile duct ligation + magnesium glycyrrhizinate injection (BMc group), common bile duct ligation + magnesium glycyrrhizinate injection high dose group BMh group); common bile duct ligation + 0.9% saline group (BN group) and sham operation group (Sham group). The BN and Sham groups were injected intraperitoneally with normal saline at 30 mg / kg / day. The BMc group was injected intraperitoneally with magnesium isoglycyrrhizinate at 30 mg / kg / day. The BMh group was intraperitoneally injected with magnesium isoglycyrrhizinate 60 mg / kg / d. At different times after surgery, blood was collected from the orbit of each rat and serum was obtained. The indexes of liver function, such as ALT, AST, DBil, were detected by automatic biochemical analyzer. Results: The levels of ALT and AST in serum of BN, BMc and BMh groups were significantly increased after bile duct ligation. With the increase of obstruction time, the serum levels of ALT and AST in BM and BMh groups were significantly lower than those in BN group (P <0.01), but there was no significant difference between the two groups. The serum levels of D-Bil in the BN, BMc and BMh groups after operation were significantly higher than those in the Sham group (P <0.01), but the levels of D-Bil did not change significantly with the prolongation of obstruction . Conclusion: Magnesium isrimazole injection can effectively reduce the level of serum aminotransferase in rats with obstructive jaundice, and has a significant protective effect on liver function.
