[目的]研究中药白花蛇舌草(Hedyotis diffusa HD)对人喉癌Hep-2细胞凋亡、周期及增殖的影响,以探讨其在喉癌肿瘤治疗方面的作用机制。[方法]分别用不同浓度的HD作用于体外培养的Hep-2细胞,MTT法检测药物对细胞增殖抑制作用,流式细胞术检测细胞凋亡和细胞周期变化。[结果]HD浓度为80mg/mL时,MTT 48h和72h细胞抑制率分别为43.80%和55.00%,与对照组比较有显著性差异(P<0.01);细胞周期结果显示随着药物浓度增加和作用时间延长,G0/G1期细胞减少,S期细胞显著性增加,细胞阻滞在S期;细胞早期凋亡率明显增加,48h和72h凋亡率分别为7.10%和19.47%,与对照组1.49%和2.75%比较结果有显著性差异(P<0.01)。[结论]HD浓度为80mg/mL时对体外培养的Hep-2细胞具有明显增殖抑制作用,使细胞阻滞在S期,并诱导细胞发生凋亡。
[Objective] To investigate the effect of Hedyotis diffusa HD on the apoptosis, cycle and proliferation of human laryngeal carcinoma Hep-2 cells in order to investigate its mechanism in the treatment of laryngeal cancer. [Methods] Hep-2 cells cultured in vitro were treated with different concentrations of HD. The inhibitory effects of drugs on cell proliferation were detected by MTT assay. Apoptosis and cell cycle changes were detected by flow cytometry. [Results] The inhibitory rates of MTT 48h and 72h at HD 80mg / mL were 43.80% and 55.00%, respectively, which were significantly different from the control group (P <0.01). The cell cycle results showed that with the increase of drug concentration and The cells in G0 / G1 phase decreased and S phase cells increased significantly, and the cells arrested in S phase. The early apoptosis rate was significantly increased, the apoptotic rate was 7.10% and 19.47% at 48h and 72h respectively. Compared with the control group There was significant difference between 1.49% and 2.75% (P <0.01). [Conclusion] HD inhibited the proliferation of Hep-2 cells cultured in vitro at a concentration of 80 mg / mL, blocked cells in S phase and induced cell apoptosis.
