本实验以形态和功能相结合的方法应用细胞培养、扫描电镜、透射电镜及放射免疫等多种技术,研究了锗-132对体外培养大鼠腺垂体细胞的作用。Ge-132作用于细胞后,放免测定细胞上清液中生长激素(GH)、催乳激素(LTH)。Ge-132浓度为0.01mmol/L组与1mmol/L组比较GH、LTH浓度增高显著;实验组促甲状腺激素(TSH)、卵泡刺激素(FSH)、黄体生成素(LH)较对照组明显降低,且随着Ge-132浓度的增高,激素TSH、PSH、LH浓度降低更为显著。电镜观察腺垂体嗜酸性细胞里分泌功能旺盛相,嗜碱性细胞呈分泌功能不活跃状态。
In this experiment, a combination of morphology and function using cell culture, scanning electron microscopy, transmission electron microscopy and radioimmunoassay, and other techniques to study the role of technetium-132 in vitro culture of rat pituitary cells. After Ge-132 was applied to the cells, radioimmunoassay of growth hormone (GH) and prolactin (LTH) in the cell supernatant was performed. The concentration of GH and LTH in Ge-132 group was significantly higher than that in 1 mmol/L group, and significantly lower in the experimental group than in the control group. With the increase of Ge-132 concentration, the concentrations of hormones TSH, PSH, and LH decreased more significantly. Electron microscopy showed that the secretory function of the eosinophils in the pituitary gland was vigorous and the basophils were inactive.
