目的:观察PDGF对鼠成骨细胞增殖能力的影响及其与BMP、bFGF联合应用对细胞的生长有否协同作用。方法:酶消化分段收集法分离培养出鼠成骨细胞,利用MTT法检测活细胞产生甲月替结晶的含量多少,反映细胞的增殖能力和生长情况。结果:PDGF能促进成骨细胞的增殖生长,其有效浓度为1~100ng/ml,以100ng/ml作用最大;PDGF与BMP、bFGF联合应用可明显增强成骨细胞的增殖能力。结论:PDGF可以刺激成骨细胞的增殖,且与BMP、bFGF有协同作用。MTT法是测定成骨细胞增殖能力较理想的方法
OBJECTIVE: To observe the effect of PDGF on the proliferation of rat osteoblasts and its synergistic effect with BMP and bFGF on the cell growth. METHODS: Rat osteoblasts were isolated and cultured by enzymatic digestion and fractionated method. MTT assay was used to determine the amount of formazan crystals produced by living cells, which reflected the proliferation and growth of cells. Results: PDGF could promote the proliferation and proliferation of osteoblasts. The effective concentration of PDGF was 1 ~ 100ng / ml, and the effect was greatest at 100ng / ml. Combination of PDGF with BMP and bFGF could significantly enhance the proliferation of osteoblasts. Conclusion: PDGF can stimulate the proliferation of osteoblasts and has synergistic effect with BMP and bFGF. MTT assay is an ideal method for the determination of osteoblast proliferation
