缺氧诱导因子(hypoxia inducible factor,HIF)是一种螺旋-环-螺旋转录因子,具有许多基因的氧敏感性,这些基因的表达与组织或细胞中氧含量密切相关,且HIF可以激活与无氧酵解及血管生成中许多基因的表达。在核因子(nuclear factor,NF)-κB信号通路中,IκB激酶(IκB kinase,IKK)与肿瘤坏死因子(tumor necrosis factor,TNF)等细胞因子也能通过不同的信号通路刺激HIF的合成。目前的研究已经表明,在缺氧性炎症中,二者对缺氧呈现出不同程度的敏感性,且具有密切的相互依赖性。本文通过阐述HIF与NF-κB在缺氧性炎症中相互依赖性调控机制,旨在为进一步研究具有缺氧症特点疾病以及为寻求治疗新靶点提供重要的参考资料。
Hypoxia inducible factor (HIF) is a helix-loop-helix transcription factor that has the oxygen sensitivity of many genes. The expression of these genes is closely related to the oxygen content in tissues or cells, and HIF can activate and Oxygen glycolysis and angiogenesis in many gene expression. In the nuclear factor (NF) -κB signaling pathway, cytokines such as IκB kinase (IKK) and tumor necrosis factor (TNF) can also stimulate the synthesis of HIF through different signaling pathways. The current research has shown that in hypoxic inflammation, both of them show different degrees of sensitivity to hypoxia, and have close interdependence. In this paper, the inter-dependent regulation of HIF and NF-κB in hypoxic inflammation is described in order to provide important references for the further study of the diseases characterized by hypoxia and the search for new targets.
