目的采用血管内超声评价短期内不同剂量阿托伐他汀对动脉粥样硬化(AS)家兔模型斑块的影响。方法 30只新西兰大白兔随机分为对照组(n=7)、模型组(n=7)、阿托伐他汀低剂量组(n=8)和阿托伐他汀高剂量组(n=8)。对照组给予普通饲料喂养,其他各组均通过8周高脂喂养联合腹主动脉球囊损伤术建立AS模型。于第8周末行血管内超声检查。8周后,模型组,阿托伐他汀低、高剂量组继续高脂喂养,分别给予低、高剂量组兔不同剂量的阿托伐他汀干预2周。10周处死所有动物。抽取兔静脉血,测定血清血脂水平,应用ELISA法测定血清超敏C反应蛋白(hs-CRP)。并行血管内超声和病理组织学检查。结果对照组血脂水平明显低于其他各组(P<0.05),阿托伐他汀低、高剂量两组血脂水平和血清hs-CRP水平明显低于模型组(P<0.05),且高剂量组变化更加明显(P<0.01)。血管内超声显示模型组的血管外弹力膜面积(EEMA)、斑块面积(PA)、管腔面积狭窄率(LAS%)明显大于低、高剂量组(P<0.05),管腔面积(LA)差异无统计学意义(P>0.05)。结论阿托伐他汀能够短时间内降低AS兔的血脂水平,并减少AS兔斑块的形成,且与药物剂量呈正相关。
Objective To evaluate the effect of different doses of atorvastatin on plaque in atherosclerosis (AS) rabbits by intravascular ultrasound in the short term. Methods Thirty New Zealand rabbits were randomly divided into control group (n = 7), model group (n = 7), atorvastatin low dose group (n = 8) and atorvastatin high dose group . The control group was fed with normal feed, and the other groups were given AS model by 8-week high-fat diet combined with abdominal aorta balloon injury. At the end of the 8th weekend, intravascular ultrasound was performed. After 8 weeks, the model group and the atorvastatin low and high dose groups continued to be fed with high fat diet. Atorvastatin with different dosages was given to the low and high dose groups respectively for 2 weeks. All animals were executed at 10 weeks. The venous blood of rabbits was drawn and the level of serum lipids was measured. The serum hs-CRP level was measured by ELISA. Parallel intravascular ultrasound and histopathology. Results The levels of lipid in control group were significantly lower than those in other groups (P <0.05). The levels of serum lipids and serum hs-CRP in atorvastatin low and high dose groups were significantly lower than those in model group (P <0.05) The change is more obvious (P <0.01). The intravascular ultrasound showed that the area of extravasated elastic membrane (EEMA), plaque area (PA) and lumen area stenosis rate (LAS%) in the model group were significantly higher than those in the low and high dose groups (P <0.05) ) Difference was not statistically significant (P> 0.05). Conclusions Atorvastatin can reduce the level of blood lipids in AS rabbits and reduce the formation of plaques in AS rabbits in a short period of time, which is positively correlated with the dose of drugs.
