目的:观察冠脉通片对大鼠急性心肌梗死治疗作用及对心肌保护作用机制。方法:采用冠状动脉左前降支结扎术制备大鼠急性心肌梗死模型,术前预防性给药3d,1次/d,术后继续给药7d,通过死亡率、心电图、血浆肌钙蛋白T、心肌梗死重量、心肌缝隙连接蛋白Cx43、心肌胶原纤维天狼星红染色等指标,阐明冠脉通片对心肌梗死的治疗作用及作用机制。结果:冠脉通大鼠冠状动脉结扎即刻死亡率均较模型对照组低,以1.3g生药/kg组最明显,死亡率为0;冠脉通各剂量组术后即刻心电图ST段变化较模型对照组明显改善,且术后7d仍较模型对照组明显改善。冠脉通2.6、1.3g生药/kg组肌钙蛋白T明显低于模型对照组。各剂量组心肌梗死重量百分比均较模型对照组明显减少。心肌Cx染色可见模型对照组Cx表达明显减少,分布紊乱,冠脉通2.6g生药/kg组较模型对照组表达明显增多。模型对照组大鼠心肌经天狼星红染色后可见明显的、大面积红色胶原染色,冠脉通2.6g生药/kg组无1例胶原染色,1.3g生药/kg组可见胶原染色,程度低于2.6g生药/kg组,0.65g生药/kg组可见红色胶原染色,程度与模型对照组接近。结论:冠脉通预防给药后,可明显降低冠状动脉结扎即刻大鼠死亡率,改善心电图ST抬高,降低心肌梗死范围,增加心肌Cx43表达,改善心肌胶原纤维化。
Objective: To observe the therapeutic effect of “Guanmaitong tablet” on acute myocardial infarction in rats and its mechanism of myocardial protection. Methods: Acute myocardial infarction model was established by ligation of left anterior descending coronary artery in rats. Preoperative prophylactic administration was given for 3 days and once daily for 7 days. The mortality, electrocardiogram, plasma Troponin T, Myocardial infarction weight, myocardial connexin Cx43, myocardial collagen fibers Sirius red staining and other indicators, to clarify the Coronary Tongliao Tablet for the treatment of myocardial infarction and its mechanism of action. Results: Immediate coronary artery occlusion rate of coronary artery was lower than that of the model control group, the most obvious was 1.3g crude drug / kg group, the mortality rate was 0. The coronary artery ST segment changes in each dose group were shorter than the model The control group improved significantly, and after 7 days than the model control group was significantly improved. Coronary artery pass 2.6, 1.3g crude drug / kg group, troponin T was significantly lower than the model control group. The weight percentage of myocardial infarction in each dose group was significantly reduced compared with model control group. Myocardial Cx staining showed that the model control group Cx expression was significantly reduced, the distribution of disorders, coronary artery through 2.6g crude drug / kg group was significantly increased compared with the model control group. The myocardium of the model control group showed obvious, large area red collagen staining after Sirius red staining. There was no collagen staining in coronary artery through 2.6g crude drug / kg group, collagen staining was observed in 1.3g crude drug / kg group, and the degree was lower than 2.6 g crude drug / kg group, 0.65g crude drug / kg group visible red collagen staining, the extent of the model control group close. CONCLUSION: Coronary prophylaxis can decrease the mortality rate of rat immediately after ligation of coronary artery, improve ST elevation, reduce the range of myocardial infarction, increase the expression of Cx43 in myocardium and improve myocardial collagen fibrosis.
