西地那非(Sidenafil,万艾可,俗称伟哥)是一种非特异性5型磷酸二酯酶抑制剂,对环鸟苷单磷酸酯合成酶(cGMP)具有抑制作用。一氧化氮释放增多可导致cGMP水平提高。其主要临床效应是促进海绵体平滑肌松弛,进入阴茎血流量增加,有助于勃起功能障碍的缓解。 较轻的副作用为头痛、潮红、鼻塞、色彩视觉缺陷和严重的心血管、脑血管副作用,均有报道,到目前为止尚未报道属于神经系统副作用的惊厥性癫痫发作。本文作者报告2例病人在服用西地那非不久,首次发生癫痫发作。
Sidenafil (Viagra, commonly known as Viagra) is a nonspecific 5-type phosphodiesterase inhibitor that inhibits cyclic guanosine monophosphate synthase (cGMP). Increased nitric oxide release leads to increased cGMP levels. The main clinical effect is to promote sponge smooth muscle relaxation, increased blood flow into the penis, contribute to the erection of erectile dysfunction. Smaller side effects are headache, flushing, stuffy nose, color vision defects and severe cardiovascular and cerebrovascular side effects. It has been reported that no seizures have been reported so far that are side effects of the nervous system. The authors report the first seizures in 2 patients shortly after taking sildenafil.
