[目的]研究Survivin、Fas在骨肉瘤组织中的表达情况与临床组织分型、外科分期的关系,初步探讨二者在骨肉瘤发生、发展中的作用、临床意义及相关性。[方法]回顾性研究潍坊市人民医院骨科2002~2010年所收治的36例骨肉瘤患者临床资料及病理标本,应用免疫组化SP法检测Survivin、Fas蛋白在人骨肉瘤组织中表达情况,并结合患者的年龄、性别、肿瘤大小、部位、转移、组织类型和Enneking分期等进行综合分析,用SPSS 17.0统计学软件对实验数据进行统计学处理,Survivin、Fas的相关性采用Spearman等级相关分析。[结果]36例骨肉瘤组织中Survivin蛋白阳性表达率为77.8%,Survivin蛋白阳性表达与患者性别、年龄、肿瘤大小、肿瘤部位、组织类型无相关性(P>0.05),而与远处转移及Enneking外科分期相关(P<0.05),Fas在36例骨肉瘤中表达的阳性率为47.22%,Fas阳性表达与患者性别、年龄、肿瘤大小、肿瘤部位、组织类型无明显相关性(P>0.05),而与远处转移及临床外科分期明显相关(P<0.05)。Suvivin蛋白阳性表达组中,Fas蛋白阳性表达率为59.26(16/27),Suvivin蛋白阴性表达组中,Fas蛋白阴性表达率为88.89%(8/9)。经Spearman等级相关分析,两者在骨肉瘤组织中表达呈正相关(r=0.418,P<0.05)。[结论]Survivin和Fas在骨肉瘤中表达具有明显的相关性,提示骨肉瘤中Survivin、Fas蛋白在骨肉瘤的发生发展中起到一定的协同作用,可作为诊断及治疗骨肉瘤的重要参考指标。
[Objective] To investigate the relationship between the expression of Survivin and Fas in osteosarcoma tissue and the clinical tissue typing and surgical staging, and to explore their roles, clinical significance and relevance in the pathogenesis and development of osteosarcoma. [Methods] The clinical data and pathological specimens of 36 patients with osteosarcoma admitted from Department of Orthopedics, Weifang People’s Hospital from 2002 to 2010 were retrospectively studied. The expression of Survivin and Fas protein in human osteosarcoma tissues was detected by immunohistochemical SP method. The patient’s age, sex, tumor size, location, metastasis, histological type and Enneking staging were analyzed comprehensively. The experimental data were statistically analyzed by SPSS 17.0 statistical software. The correlation between Survivin and Fas was analyzed by Spearman rank correlation analysis. [Results] The positive rate of Survivin protein expression in 77 osteosarcoma tissues was 77.8%. The positive expression of Survivin protein had no correlation with the gender, age, tumor size, tumor location and type (P> 0.05) (P <0.05). The positive expression rate of Fas in 36 cases of osteosarcoma was 47.22%. There was no significant correlation between the positive expression of Fas and sex, age, tumor size, tumor location and type of tissue (P> 0.05), but significantly correlated with distant metastasis and clinical staging (P <0.05). The positive expression rate of Fas protein in Suvivin protein positive group was 59.26 (16/27), while the negative expression rate of Fas protein in Suvivin protein negative group was 88.89% (8/9). Spearman rank correlation analysis showed that there was a positive correlation between them in osteosarcoma (r = 0.418, P <0.05). [Conclusion] The expression of Survivin and Fas in osteosarcoma has a significant correlation, suggesting that Survivin and Fas protein in osteosarcoma play a synergistic role in the occurrence and development of osteosarcoma and can be used as an important reference for diagnosis and treatment of osteosarcoma .
