目的:探讨羊栖菜多糖提取物(SFPS)对阿尔茨海默病(AD)大鼠模型行为的干预作用及脑皮质Bcl-2和Bax基因表达的影响。方法:制作D-半乳糖阿尔茨海默病大鼠模型,设计正常对照组、模型组、吡拉西坦片、羊栖菜多糖提取物不同剂量组,观察大鼠行为学及脑皮质Bcl-2和Bax基因表达指标的改变。结果:与正常对照组相比,模型组学习记忆能力显著下降(P<0.05),其Bcl-2/Bax值下降;与模型组相比,0.8g/kg、1.6g/kg羊栖菜多糖提取物治疗组均能较好的改善学习记忆能力,且具有一定剂量依赖性,其Bcl-2/Bax值增加。结论:SFPS能调节海马组织Bcl-2和Bax的表达,显著提高Bcl-2/Bax值,抑制海马神经元的凋亡,改善AD大鼠学习记忆能力。
Objective: To investigate the effect of SFPS on the behavior of Alzheimer’s disease (AD) rat model and the expression of Bcl-2 and Bax gene in cerebral cortex. Methods: The D-galactose-induced Alzheimer’s disease rat model was made. The normal control group, the model group, the piracetam tablet and the polysaccharide extract of Sargassum fusiforme were given in different dosage groups. The behavior and the expression of Bcl- 2 and Bax gene expression changes. Results: Compared with the normal control group, the learning and memory abilities of the model group decreased significantly (P <0.05), and the Bcl-2 / Bax values of the model group decreased. Compared with the model group, 0.8g / kg, 1.6g / The extract treatment group can better improve learning and memory ability, and with a dose-dependent, the Bcl-2 / Bax value increased. Conclusion: SFPS can regulate the expression of Bcl-2 and Bax in hippocampus, significantly increase the Bcl-2 / Bax, inhibit the apoptosis of hippocampal neurons and improve the learning and memory ability of AD rats.
