琼脂双扩散血清试验表明,参试的我国花生矮化病毒(PSV)6个分离物和株系之间血清学关系十分亲近,而与美国PSV-E和PSV-W株系存在明显的差异。完成对花生致病力不同的PSV-Mi和PSV-S两个株系壳蛋白(CP)基因的RT-PCR扩增、克隆和序列分析。两个株系CP基因均含654个核苷酸,编码218个氨基酸。两株系之间CP基因核苷酸序列同源性为99.1%,推导的CP氨基酸序列同源性为99.5%。与已报道的PSV亚组IPSV-ER和PSV-J株系CP基因核苷酸序列同源性为75.7%~77.8%,与PSV亚组ⅡPSV-WCP基因核苷酸序列同源性为74.3%-74.6%。鉴于我国FSV株系血清学性质和CP基因核苷酸序列同源性与PSV两个亚组的PSV株系存在明显的差异,建议将我国PSV株系划为一个新的PSV亚组,即PSV亚组-Ⅲ。
The results of agar double-diffusion assay showed that the serological relationships among the six isolates of peanut dwarf virus (PSV) tested in our country were very close to each other, but significantly different from the strains of PSV-E and PSV-W in the United States. Complementation of the RT-PCR amplification, cloning and sequence analysis of the capsid (CP) genes of two strains of PSV-Mi and PSV-S with different pathogenicity. Both strains of CP gene contain 654 nucleotides, encoding 218 amino acids. The CP gene nucleotide sequence homology between the two strains was 99.1%, and the deduced CP amino acid sequence homology was 99.5%. The homology of nucleotide sequence of CP gene to IPSV-ER and PSV-J strains reported in PSV subgroup was 75.7% -77.8%, which was homologous with PSV subgroup Ⅱ PSV-WCP gene nucleotide sequence Sex is 74.3% -74.6%. In view of the significant differences in the serological characteristics and CP gene nucleotide sequence homology between FSV strains and PSV strains in our two subgroups, it is suggested that PSV strains in our country should be classified as a new PSV subgroup, namely PSV Subgroup-III.
