目的:探讨益气活血化痰方作用的分子机制,为临床益气活血化痰方早期介入治疗中风病选取最佳的时间点。方法:健康Wistar大鼠,雌雄各半80只,随机分为空白对照组、假手术组、再灌注3h组、再灌注24h组、再灌注3d组、再灌注3h治疗组、再灌注24h治疗组、再灌注3d治疗组共8组,并在3h、24h、72h分别用益气活血化痰方灌胃,观察不同时间点对GLUT-1及 ICAM-1的影响。结果:益气活血化痰方能增加脑缺血时GLUT-1的表达,降低ICAM-1的表达;3h时间点使用益气活血化痰方对GLUT-1、ICAM-1表达的改变影响最大。结论:增加缺血时GLUT-1表达、降低ICAM-1表达是益气活血化痰方治疗缺血性中风作用的分子机制之一;早期(再灌注3h)使用中药效果最佳。
Objective: To explore the molecular mechanisms of the effects of Qi-invigorating, blood-activating and stasis-resolving prescriptions and to select the optimal time point for the early interventional treatment of apoplexy with Yiqihuoxuehuayu prescription. METHODS: Healthy Wistar rats, 80 males and 80 females, were randomly divided into blank control group, sham operation group, reperfusion 3 h group, reperfusion 24 h group, reperfusion 3 d group, reperfusion 3 h treatment group, and reperfusion 24 h treatment group. The rats were treated with Yiqi Huoxue Huayu Decoction, and the effects of different time points on GLUT-1 and ICAM-1 were observed in 3 groups treated with reperfusion for 8 days. Results: Yiqihuoxuehuayu prescription can increase the expression of GLUT-1 and decrease the expression of ICAM-1 during cerebral ischemia. The effect of Yiqihuoxuehuayu prescription on the changes of GLUT-1 and ICAM-1 expression is greatest at the 3h time point. . Conclusion: Increasing the expression of GLUT-1 and decreasing the expression of ICAM-1 during ischemia is one of the molecular mechanisms of Yiqi Huoxue Huayu Recipe in the treatment of ischemic stroke. Early (3h after reperfusion) Chinese medicine has the best effect.
