目的全面了解Matrilysin在胃癌组织中的表达情况,分析其与胃癌侵袭、转移和预后的相关性。方法以52例胃癌患者为研究对象,分别应用RT-PCR、Western blot及免疫组织化学方法检测Matrilysin在胃原发癌、胃正常黏膜及淋巴结转移癌组织中的表达,对所有病例进行随访,分析Matrilysin与胃癌侵袭、转移和预后的相关性。结果 Matrilysin在胃原发癌和淋巴结转移癌组织中表达明显增高,而在胃正常黏膜组织中表达较弱或缺失,差异有统计学意义(P<0.05);其mRNA和蛋白质表达水平具有较高的一致性(P<0.001)。Matrilysin表达在胃原发癌为浸润型癌、癌侵透浆膜、浆膜受侵面积>20cm2、淋巴结分期晚者(N3)中明显增高(P<0.001)。Matrilysin高表达组患者5年生存率为34.1%,而Matrilysin低表达组为55.6%,差异有统计学意义(χ2=9.778,P=0.002)。结论 Matrilysin基因表达与胃癌的侵袭、转移和预后有关,是反映胃癌恶性生物学行为较好的分子指标。
Objective To comprehensively understand the expression of Matrilysin in gastric cancer and analyze its correlation with invasion, metastasis and prognosis of gastric cancer. Methods Fifty-two patients with gastric cancer were studied. RT-PCR, Western blot and immunohistochemistry were used to detect the expression of Matrilysin in primary gastric cancer, normal gastric mucosa and lymph node metastasis. All cases were followed up and analyzed Correlation of Matrilysin with Invasion, Metastasis and Prognosis of Gastric Cancer. Results The expression of Matrilysin was significantly higher in primary gastric cancer and lymph node metastasis than in normal gastric mucosa (P <0.05), and higher in mRNA and protein expression Consistency (P <0.001). Matrilysin expression in invasive primary gastric cancer, cancer invasion through the serosa, serosal invasion area> 20cm2, lymph node staging were significantly higher (P <0.001). The 5-year survival rate of Matrilysin high expression group was 34.1%, while Matrilysin low expression group 55.6%, the difference was statistically significant (χ2 = 9.778, P = 0.002). Conclusion Matrilysin gene expression is related to the invasion, metastasis and prognosis of gastric cancer, which is a good molecular indicator of malignant behavior of gastric cancer.
