目的观察扶肾降浊方对系膜增生性肾炎大鼠肾小管间质细胞外基质Ⅰ、Ⅲ型胶原(COLⅠ及COLⅢ)蛋白和基因的调节作用。方法采用系膜增生性肾炎模型,将大鼠随机分为正常组、模型组、扶肾降浊方中药组、贝那普利西药组。采用免疫组化法和实时荧光定量PCR法分别检测实验12周、16周和20周末各组Ⅰ、Ⅲ型胶原蛋白和基因的表达。结果 12周、16周和20周末的模型组肾脏COLⅠ及COLⅢ蛋白表达呈强阳性(P<0.01),COLⅠ及COLⅢ基因表达上调(P<0.01),扶肾降浊方中药组和贝那普利组大鼠肾脏COLⅠ及COLⅢ蛋白表达均较模型组降低(P<0.01),COLⅠ及COLⅢ基因表达亦均较模型组下调(P<0.05);随着造模时间的延长,模型组肾小管COLⅠ及COLⅢ蛋白和基因表达均呈升高趋势,治疗组仍作用明显(P<0.05)。结论扶肾降浊方可通过基因和蛋白水平调节COLⅠ和COLⅢ在肾脏的表达,减少COLⅠ和COLⅢ在肾小管间质的积聚以减轻肾间质纤维化,从而对肾小管间质损害起到防治作用。
Objective To observe the regulatory effect of “Fu Shen Jiang Zhuo Fang” on the protein and gene expression of type Ⅰ and type Ⅲ collagen (COL Ⅰ and COL Ⅲ) in mesangial proliferative glomerulonephritis rats. Methods Mesangial proliferative glomerulonephritis model was used. The rats were randomly divided into normal group, model group, traditional Chinese medicine group of Fuzheng Jiangzhuo Decoction and benazepril western medicine group. Immunohistochemistry and real-time fluorescence quantitative PCR were used to detect the expression of type Ⅰ and type Ⅲ collagen and gene in each group at 12, 16 and 20 weeks after the experiment. Results The expressions of COL Ⅰ and COL Ⅲ protein in the kidney of model group at 12 weeks, 16 weeks and 20 weeks were strongly positive (P <0.01) and COL Ⅰ and COL Ⅲ genes were up-regulated (P <0.01). Fuzheng Jiangzhuo Decoction and benazepril Compared with the model group, the expression of COL Ⅰ and COL Ⅲ protein in the kidney of the model group was significantly lower than that of the model group (P <0.01), and the expression of COL Ⅰ and COL Ⅲ gene were also decreased compared with the model group (P <0.05). With the extension of modeling time, COL Ⅰ and COL Ⅲ protein and gene expression showed an upward trend in the treatment group was still significant (P <0.05). Conclusion Fuzhengjiangzhuo can regulate the expression of COL Ⅰ and COL Ⅲ in the kidney through gene and protein level and reduce the accumulation of COL Ⅰ and COL Ⅲ in the tubulointerstitium so as to relieve the renal interstitial fibrosis and thus prevent the tubulointerstitial damage effect.
