目的探讨TGF-β1基因作用于平滑肌细胞从而促进内皮细胞黏附的作用。方法用单价阳离子脂质体转染试剂DOTAP转染pMAMneo TGF-β1于原代培养的平滑肌细胞,经G418筛选。检测转染pMAMneo TGF-β1和转染pMAMneo的平滑肌细胞黏附内皮细胞的情况。结果经G418筛选,转染pMAMneo TGF-β1组转化生长因子TGF-β1基因在平滑肌细胞得到有效表达,内皮黏附实验表明转染pMAMneo TGF-β1组平滑肌细胞表面有大量内皮细胞黏附[(32±2)/高倍视野],而对照组平滑肌细胞仅有少量内皮细胞黏附[(11±1)/高倍视野],其与转染pMAMneo组相比,差异显著(P<0.01)。结论 TGF-β1基因能有效作用于血管平滑肌细胞从而促进内皮细胞黏附,在血管再生中具有重要作用。
Objective To investigate the effect of TGF-β1 gene on smooth muscle cells and promote the adhesion of endothelial cells. Methods The primary cultured human smooth muscle cells were transfected with pMAMneo TGF-β1 by monovalent cationic liposome transfection reagent DOTAP, and then selected by G418. The adhesion of endothelial cells to smooth muscle cells transfected with pMAMneo TGF-β1 and pMAMneo was detected. Results After transfected with pMAMneo TGF-β1, the TGF-β1 expression in the transfected pMAMneo TGF-β1 group was significantly increased in smooth muscle cells. The endothelial adhesion assay showed that a large number of endothelial cells adhered to the smooth muscle cells transfected with pMAMneo TGF-β1 [(32 ± 2 ) / High power field], while the control group had only a small amount of endothelial cell adhesion [(11 ± 1) / high power field], which was significantly different from that of pMAMneo group (P <0.01). Conclusion TGF-β1 gene can effectively act on vascular smooth muscle cells and promote endothelial cell adhesion, which plays an important role in angiogenesis.
