[目的]同时测定葎草中木犀草素-7-O-β-D-葡萄糖苷和大波斯菊苷的含量。[方法]采用高效液相色谱法(HPLC)测定葎草中木犀草素-7-O-β-D-葡萄糖苷和大波斯菊苷的含量,色谱柱kromasilC18,以甲醇-0.2%磷酸溶液(45:55)为流动相,流速为1.0mL/min,柱温35℃,检测波长为350nm。[结果]木犀草素-7-O-β-D-葡萄糖苷在0.072~0.36μg的范围内与峰面积积分值成良好的线性关系(r=0.9999),精密度相对平均标准差(RSD)=2.06%,平均加样回收率为99.3%,RSD为2.8%;大波斯菊苷在0.05~0.40μg的范围内与峰面积积分值成良好的线性关系(r=0.9997),精密度RSD=1.73%,平均加样回收率为100.2%,RSD%为2.7%。葎草中木犀草素-7-O-β-D-葡萄糖苷的含量在7、8月份含量最高,大波斯菊苷在6月份含量最高。[结论]该方法简单、灵敏、稳定、可靠,可用于葎草中木犀草素-7-O-β-D-葡萄糖苷和大波斯菊苷的含量测定。葎草适宜的采收季节应在6~8月份。
[Objective] The aim was to determine the content of luteolin-7-O-β-D-glucoside and cosmosin at the same time. [Method] The content of luteolin-7-O-β-D-glucoside and macro- cosmosin in Pteris vittata was determined by high performance liquid chromatography (HPLC). The column kromasil C18 was prepared by methanol-0.2% phosphoric acid solution ( 45:55) as mobile phase, flow rate 1.0 mL/min, column temperature 35°C, detection wavelength 350 nm. [Results] The linearity of luteolin-7-O-β-D-glucoside in the range of 0.072-0.36 μg was in good linear relationship with the integrated value of peak area (r=0.9999). The relative standard deviation of precision (RSD) =2.06%, the average recovery was 99.3%, the RSD was 2.8%; the crestulose was in a good linear relationship with the integrated value of the peak area in the range of 0.05-0.40 μg (r = 0.9997), precision RSD = At 1.73%, the average recovery was 100.2%, and the RSD% was 2.7%. The content of luteolin-7-O-β-D-glucoside in Valerian had the highest content in July and August, while the content of luteolin was the highest in June. [Conclusion] This method is simple, sensitive, stable and reliable. It can be used for the determination of luteolin-7-O-β-D-glucoside and cosmosin in Radix hepatica. The suitable harvesting season for barnyardgrass should be from June to August.
