目的建立同时测定清热除湿汤中龙胆苦苷和黄芩苷含量的高效液相色谱(HPLC)测定方法。方法采用Diamonsil C18色谱柱(250 mm×4.6 mm,5μm);甲醇-0.2%磷酸梯度洗脱,流速:1 m L/min;检测波长:270 nm。结果龙胆苦苷和黄芩苷分别在0.225 2~2.252 0μg(r=1,n=5)和0.246 0~2.460 0μg(r=0.999 9,n=5)范围内有良好的线性关系,平均加样回收率分别为100.12%和98.38%(n=6)。结论本法简单、快速、准确,可用于清热除湿汤的质量控制。
Objective To establish a high performance liquid chromatography (HPLC) method for the simultaneous determination of gentiopicroside and baicalin in Qingre Decoction. Methods Diamonsil C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase was eluted with methanol - 0.2% phosphoric acid. The flow rate was 1 m L / min. The detection wavelength was 270 nm. Results There was a good linear relationship between gentiopicroside and baicalin in the range of 0.225 2 ~ 2.252 0 μg (r = 1, n = 5) and 0.246 0 ~ 2.460 0 μg (r = 0.999 9, n = 5) The recoveries were 100.12% and 98.38%, respectively (n = 6). Conclusion This method is simple, rapid and accurate and can be used for quality control of Qingre Decoction.
