目的 :探讨肿瘤坏死因子 (TNF)、白细胞介素 8(IL 8)在脂多糖 (lipopolysaccharide ,LPS)致大鼠脑水肿发生中的变化及作用。方法 :LPS诱发大鼠脑水肿发生后 ,制备脑组织匀浆 ,ELISA法测定IL 8含量 ,放免法测定TNF α含量。结果 :LPS注入后TNF α、IL 8相继释放增加 ,与A组、NS组比较差异有非常显著性 (P <0 0 1 ) ,48h后仍维持较高水平 ,LPS组脑组织含水量与Evansblue(EB)含量 ,TNF α与脑组织含水量 ,IL 8与脑组织含水量 ,IL 8与EB含量均呈正相关 ,r值分别为 0 537、0 42 3、0 473、0 831。P <0 0 5。结论 :TNF α、IL 8与脑组织炎性损伤密切相关 ,介导了脂多糖导致脑水肿的发生、发展过程
Objective: To investigate the changes of tumor necrosis factor (TNF) and interleukin 8 (IL 8) in the development of cerebral edema induced by lipopolysaccharide (LPS) in rats. Methods: After brain edema induced by LPS in rats, homogenate of brain tissue was prepared. The content of IL 8 was determined by ELISA, and the content of TNFα was determined by radioimmunoassay. RESULTS: After LPS injection, the release of TNFα and IL-8 were significantly increased (P <0.01), and the levels of TNFα and IL-8 in the LPS group were still significantly higher than those in the Evansblue group (EB), TNFα and brain water content, IL 8 and brain water content, IL 8 and EB content were positively correlated, r values were 0 537,0 42 3,0 473,0 831. P <0 0 5. CONCLUSION: TNFα and IL-8 are closely related to the inflammatory injury of brain tissue and mediate the occurrence and development of cerebral edema caused by lipopolysaccharide
