目的:探讨复色2号方治疗白癜风的分子学作用机制。方法:采用MTT法测定复色2号方对B16细胞增值的影响,在酶标仪下测定并记录B16细胞增殖情况;采用酪氨酸酶多巴速率氧化法测定复色2号方对酪氨酸酶活性的影响;采用NaOH裂解法测定B16细胞黑素含量。结果:复色2号方药物浓度在1~1000g/L对黑素瘤B16细胞增殖无统计学意义(P>0.05),但对酪氨酸酶活性和黑素合成有统计学意义(P<0.05),呈浓度依赖性。结论:复色2号方对黑素瘤B16细胞增殖无影响,但对酪氨酸酶活性和黑素合成有较强的剂量相关性促进作用,可以达到治疗白癜风的目的。
Objective: To explore the molecular mechanism of Fucai No.2 prescription in the treatment of vitiligo. Methods: MTT method was used to determine the effect of Fuse No.2 on the proliferation of B16 cells. The proliferation of B16 cells was measured and recorded under the microplate reader. The tyrosinase Doppler rate was used to determine the effect of Fuse No.2 on tyrosinase The enzyme activity was measured. The content of melanin in B16 cells was determined by NaOH cleavage method. Results: The results showed that there was no significant difference in the proliferation of melanoma B16 cells between 1 and 1000 g / L (P> 0.05), but the tyrosinase activity and melanin synthesis were statistically significant (P < 0.05), in a concentration-dependent manner. CONCLUSION: Fucai No.2 has no effect on the proliferation of melanoma B16 cells, but it has a strong dose-dependent promotion on tyrosinase activity and melanin synthesis, which can achieve the goal of treating vitiligo.
