探讨PMN-弹力蛋白酶抑制剂MDL27 324对急性胰腺炎病情演变的影响。材料和方法:将40只SD大鼠随机分成:A组,PMN-弹力蛋白酶处理组;B组,单纯诱模组;C组,MDL27 324处理组;D组,假手术组。各组动物6h后处死。取胰腺标本进行病理检查,采用Schmidt方法进行半定量分级。结果:A、B、C各组胰腺组织有不同程度的胰腺腺泡坏死、炎性细胞浸润和间质出血,程度依次减轻,其中A组炎性细胞浸润和间质出血十分显著。结论:过度激活的粒细胞在胰腺组织中浸润并大量释放的PMN-弹力蛋白酶是胰腺出血坏死不断发展的关键因素,MDL27 324具有减轻胰腺炎病理损害的作用。
To investigate the effect of PMN-elastase inhibitor MDL27 324 on the progression of acute pancreatitis. MATERIALS AND METHODS: Forty SD rats were randomly divided into: group A, PMN-elastase group; group B, simple model group; group C, MDL27 324 treatment group; group D, sham operation group. Animals in each group were sacrificed 6 hours later. Pancreatic specimens were taken for pathological examination, semi-quantitative grading by Schmidt method. Results: Pancreatic acinar necrosis, inflammatory cell infiltration and interstitial hemorrhage were observed in pancreatic tissues of A, B and C groups in varying degrees, and the severity of inflammatory cell infiltration and interstitial hemorrhage in group A were significant. CONCLUSIONS: PMN-elastase, an over-activated granulocyte infiltrating into pancreas and releasing in large quantities, is a key factor in the continued development of pancreatic hemorrhage and necrosis. MDL27 324 has the effect of reducing the pathological damage of pancreatitis.
