以铁皮石斛原球茎为试材,应用3L气升式生物反应器,研究了反应器不同培养时期、通气方式和接种量对铁皮石斛原球茎生物量及有效物质积累的影响,并比较了铁皮石斛不同来源材料的多糖与生物碱含量。结果表明:铁皮石斛原球茎的生物量及多糖和生物碱的积累量均在培养至30d时达到最大值,在反应器培养过程中逐步调节通气量由0.1vvm至0.3vvm,可提高原球茎的生物量及有效物质的含量,接种量为20~25g/L均可获得较高的原球茎生物量且有效物质含量和产量也较高,铁皮石斛原球茎的多糖和生物碱含量均高于1年生植株和组培苗。因此,铁皮石斛原球茎反应器培养30d,培养期间将通气量由0.1vvm调节至0.3vvm,接种量为20~25g/L有利于大量生产原球茎及其有效物质。
