几种因素对反应器铁皮石斛原球茎生长及有效物质的影响

以铁皮石斛原球茎为试材,应用3L气升式生物反应器,研究了反应器不同培养时期、通气方式和接种量对铁皮石斛原球茎生物量及有效物质积累的影响,并比较了铁皮石斛不同来源材料的多糖与生物碱含量。结果表明:铁皮石斛原球茎的生物量及多糖和生物碱的积累量均在培养至30d时达到最大值,在反应器培养过程中逐步调节通气量由0.1vvm至0.3vvm,可提高原球茎的生物量及有效物质的含量,接种量为20~25g/L均可获得较高的原球茎生物量且有效物质含量和产量也较高,铁皮石斛原球茎的多糖和生物碱含量均高于1年生植株和组培苗。因此,铁皮石斛原球茎反应器培养30d,培养期间将通气量由0.1vvm调节至0.3vvm,接种量为20~25g/L有利于大量生产原球茎及其有效物质。 Using protoplast from Dendrobium officinale as test material, the effects of aeration and inoculation amount on the biomass and active substance accumulation of protocorm-like bodies of Dendrobium candidum were studied by using 3L gas-lift bioreactor. Therefore, the protocorm-like protocorm-like bodies of Dendrobium candidum were cultured for 30 days, the aeration volume was adjusted from 0.1 vvm to 0.3 vvm during culture, and the inoculum size was 20-25 g / L, which was beneficial to the mass production of protocorm and its effective substance.
