用胶原酶消化的方法分离大鼠颅盖骨骨细胞,该细胞可形成一种骨样基质,在体外有β-磷酸甘油存在的条件下,不到两星期即可发生矿化。这一研究结果显示这种矿化大鼠成骨细胞样细胞培养系统可合成Ⅰ型、Ⅴ型、微量Ⅲ型胶原、骨钙素和骨ALP同工酶。在骨小结形成时可以检测到培养基中的骨钙素,且矿化时数值进一步提高。在矿化阶段ALP活性升高,同时还存在另一种形式的ALP,因而这种培养物可以作为研究矿化过程中骨特异性蛋白功能的一个非常有用的模型。
The rat calvarial osteoblasts were isolated by collagenase digestion and the cells formed a bone-like matrix that mineralized in less than two weeks in the presence of β-phosphoglycerol in vitro. The results of this study show that this mineralized rat osteoblast-like cell culture system can synthesize type I, type V, type III collagen, osteocalcin and bone ALP isozymes. Osteocalcin can be detected in the formation of bone nodules, and mineralization values are further improved. ALP activity is elevated during mineralization and another form of ALP is present and this culture can therefore be used as a very useful model for studying the function of bone-specific proteins in mineralization.
