目的:饮食不节可能会影响人体生化指标异常,采用动物长期高嘌呤饲料喂养法,以期得到模拟人类不良生活方式诱导的高血压动物模型。方法:采用SD大鼠长期高嘌呤饲料(普通饲料、酵母干粉、腺嘌呤按一定配比制成)喂养,每天正常饮水,连续30周建立接近人类病症的高血压大鼠模型。于造模0、8、15、17、21、30周分别采用无创尾动脉法测定大鼠收缩压(SBP)、舒张压(DBP)、平均动脉压(MBP);于8、10、12、18、20、24周取血,测定血清丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)、γ-谷氨酰转肽酶(GGT)、碱性磷酸酶(ALP)、总胆固醇(TC)、甘油三酯(TG)、尿酸(UA)、肌酐(Cr)水平;于7、9、11、16、19、22周进行尿代谢,测尿Cr、UA水平。结果:大鼠高嘌呤饲料喂养8周时,以收缩压为主,血压开始明显升高,17周时收缩压接近达到160mmHg;大鼠高嘌呤饲料喂养18周时血清ALT、AST、ALP、GGT、UA、Cr水平升高;11周时尿液UA水平降低;16周时尿液Cr水平降低,提示肝肾功能生化指标出现紊乱。实验中大鼠血清TC、TG未有显著差异,提示长期高嘌呤饲料喂养对大鼠血脂可能未有影响。结论:采用长期高嘌呤饲料喂养造模法,连续造模17周以上能使SD大鼠的血压升高到一定程度,达到轻、中度高血压水平,同时还能造成肝肾损伤等伴随症状。
OBJECTIVE: The unhealthy diet may affect the biochemical abnormalities in human body. Animal long-term high-purine feed-feeding method is used to obtain the animal model of hypertension induced by simulated unhealthy lifestyles. Methods: SD rat long-term high-purine feed (normal feed, yeast powder, adenine made at a certain ratio) fed daily normal drinking water for 30 weeks to establish close to human disorders in hypertensive rat model. The systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean arterial pressure (MBP) were measured by noninvasive tail artery at 0, 8, 15, 17, Blood samples were taken at 18, 20 and 24 weeks of age to determine serum ALT, AST, GGT, ALP, total cholesterol (TC), triglyceride (TG), uric acid (UA) and creatinine (Cr). Urine metabolism was measured at 7th, 9th, 11th, 16th, 19th, Results: At the 8th week of high purine diet, the systolic pressure was the main factor, the blood pressure began to increase significantly, and the systolic blood pressure reached 160mmHg at 17 weeks. The levels of ALT, AST, ALP, GGT , UA, Cr levels increased; urine UA levels decreased at 11 weeks; Urine Cr levels decreased at 16 weeks, suggesting that disorders of liver and kidney biochemical indicators. In the experiment, there was no significant difference in serum TC, TG, suggesting that long-term high purine feed may have no effect on blood lipids in rats. Conclusion: The long-term high-purine feed-feeding modeling method, continuous modeling for more than 17 weeks can make SD rats blood pressure increased to a certain extent, to light and moderate levels of hypertension, but also cause liver and kidney damage and other accompanying symptoms .
